摘要
目的 探讨曲尼司特 (Tranilast)对单侧输尿管梗阻 (UUO)大鼠肾间质纤维化的保护作用及机制。方法 将3 6只大鼠行右侧输尿管结扎 ,另 18只行假手术 (SIAM) ,结扎后大鼠随机分成模型组 (UUO)组和药物组 (Tranilast) ,用药组于术前一天开始给予Tranilast 2 0 0mg·kg-1·d-1,术后第 5、10及 15天分别处死各组大鼠中的 6只 ,用免疫组化方法测定肾间质转化生长因子 β(TGF -β) ,α 平滑肌动蛋白 (α SMA)及Ⅰ型 ,Ⅲ型胶原 (coIⅠ .coIⅢ )的表达并行HE、MASSON染色 ,并观察肾脏病理变化。结果 Tranilast可显著减少TGF -β ,α SMA ,coIⅠ ,coIⅢ的表达 (P <0 0 5 ) ,有效地改善肾脏的病理学改变。结论 曲尼司特抑制输尿管梗阻后肌成纤维细胞 (MyoF)释放TGF -β而调节胶原的合成 ,减少肾间质Ⅰ型 ,Ⅲ型胶原的沉积 ,从而改善UUO所致肾损伤 。
Objective To study the protective effects and mechanism of tranilast on the process of renal instertitial fibrosis following unilateral obstruction(UUO)in rats. Methods 36 Sprague-Dawley rats underwent UUO and then received vehicle(n=18) or tranilast(200mg·kg -1·d -1)was administered to rats one day before surgery and each day then after,additional 18 rats were sham operated,kidney of six rats of each group were harvested respectively at 5、10、15days after surgery.Immunohistochemistry was performed on renal tissue for transforming growth factor β(TGF-β)、α-smooth muscle actin(α-SMA).collagen Ⅰ、Ⅲ(coIⅠ、coIⅢ)and histological changes were also observed by HE and Masson stain.Results At every time point,TGF-β in tubules interstitium as well as α-SMA and coIⅠ colⅢdeposition were all significantly decreased by tranilast treatment.Tranilast also ameliorated the histological change of UUO rats. Conclusion Tranilast significantly decrease tissue TGF-beta released by myofrodlast.Resultin reducing the coI Ⅰ、coIⅢ deposition,and prevents renal interstitial fibrosis damage in unilateral ureteral odstruction
