摘要
目的研究低浓度乙醇对肝癌细胞增殖的影响及作用机理。方法采用MTT比色法观察乙醇对细胞增殖的影响;采用流式细胞仪法观察乙醇对肝癌细胞内活性氧的影响;采用琼脂凝胶电泳法观察DNA片段的梯形条带。结果低浓度乙醇抑制了细胞的增殖;乙醇作用于细胞后,在2h内引起活性氧的产生;24h后细胞出现典型的凋亡表现。结论乙醇能通过诱导细胞内活性氧的产生而引起细胞凋亡,呈明显的剂量依赖性。
Objective To study the mechanism of ethanol in low concentration on proliferation of the hepatocellular carcinoma (HCC) cells. Methods MTT colorimetry was used to evaluate the influence of ethanol on proliferation of cells. Flow cytometry was used to test reactive oxygen species (ROS) levels in cells. Agarsoe gel electrophoresis was used to assess the DNA laddering. Results Low concentration of ethanol suppressed proliferation of HCC Cells. ROS generation was obviously increased after the cells incubated in ethanol in 2 hrs. Typical DNA ladder was observed after the cells incubated in ethanol for 24 hrs. Conclusion Low concentration of ethanol can induce the apoptosis of HCC cells by generation of ROS in a dose-dependent manner.
