摘要
利用筛选出的19个随机引物对云南荞麦资源的9个种、1个变种、2个亚种的26份材料进行RAPD扩增反应,共获得162条DNA扩增带,其中多态性带153条,多态性带的比率平均为94.8%。Nei氏相似性系数的分析结果,荞麦种间平均相似系数为0.411,种内平均相似系数为0.786,表明云南荞麦资源种间比种内具有更丰富的多样性;聚类分析的结果,26份供试资源聚为3大类群:第Ⅰ大类群是小粒种类群,第Ⅱ大类群是甜荞类群,第Ⅲ大类群是苦荞类群,结合3大类群的特点,可以认为F.cymosum与F.esculentum的亲缘关系比与F.tataricum的更近;F.tataricum和F.tataricumssp.potanini与供试的其它荞麦资源之间的亲缘关系比较远;红花F.urophyllum与白花F.urophyllum之间的亲缘关系也比较远,红花F.urophyllum很可能是一个新的生态类型。
The 26 samples belong to 9 species?1 variation and 2 subspecies of wild buckwheat resources from Yunnan province were used to amplify DNA fragments with 19 RAPD primers, A total of 162 DNA bands were defected, of 153 were polymorphic. The average rate of polymorphic bands was 94.8%. The result of Nei's similar coefficient showed that the average similar coefficient between species was 0.411 while that within species 0.786. It showed that the wild buckwheat resources between species from Yunnan have more abundant genetic diverse than that within species; the result of clustering analysis showed that 26 different wild buckwheat resources were divided into three big groups:Ⅰ?small achene species group; ie Ⅱ?F.esculentum group; Ⅲ?F.tataricum group. Based on the characteristics of the three big groups, it might be that F.cymosum is closer genetic relative to F.esculentum than to F.tataricum; Either F.tataricum or F.tataricum ssp. Potanini is farther to the rest buckwheat wild species; The relationships of the red flower F.urophyllum and the white flower F.urophyllum is farther. The red flower F.urophyllum may be a new ecotype.
