东南沿海解藻酸弧菌伤口感染能力研究 被引量:9

Pathogenicity of Vibrio alginolyticus Derived from the Southeast Coastal Area of China

摘要 目的 研究从沿海某地海水中分离出的解藻酸弧菌的致病性。方法 采用毒力实验组 :35只二级昆明小鼠随机分为 4组 :实验组腹腔注射解藻酸弧菌 ,阳性对照组腹腔注射标准金黄色葡萄球菌、标准大肠埃希菌 ,阴性对照组腹腔注射无菌生理盐水 ;伤口感染实验组 :35只SPF小鼠 ,腿部致伤后随机分为 4组 :实验组 (浸泡含有细菌的人工海水 )和阳性对照组 (含有标准金黄色葡萄球菌、标准大肠埃希菌 ) ,阴性对照组 (浸泡无菌人工海水 ) ;观察小鼠一般状况、血常规、血培养、脏器培养 ,96h后活杀取脏器和伤肢病理检查。结果 毒力实验 :实验组 2 0只鼠中 4只 (2 0 % )的肝、肺有细菌生长 ;伤口感染实验 :伤口感染阳性率为 10 0 % ,活杀后脏器细菌培养阳性率为30 % ;病理学检查伤肢皮下组织大量中性粒细胞浸润、肌纤维破坏、脓肿形成。结论 自沿海某海域分离出的解藻酸弧菌在 10 6CFU/ml时 ,具有较强的感染能力 ,可致实验鼠伤口感染。 OBJECTIVE To study the pathogenicity of Vibrio alginolyticus dirived from the sea water in the southeast coastal area of China. METHODS 1. Virulence experiment: a group of 35 mice were divided into 4 groups: an experimental group with injection of V. alginolyticus into abdominal cavity, two positive control groups with injection of standard Staphylococcus aureus and Escherichia coli, separatedly into abdominal cavity and a negative control group with injection of aseptic physiological saline into the same position. 2. Wound infection experiment: a group of 35 SPF mice wounded on the legs were randomly divided into 4 groups: an experimental group (soaked in man-made sea water containing bacteria) and two positive control groups (standard S. aureus, standard E. coli, separately), and a negative control group ( soaked in aseptic man-made sea water ). general state,routine blood test, blood culture, organ culture, pathological test of the organs and wounded legs of sacrificed mice after 96 hours, and biochemical identification of the bacteria strains separated from the mice before and after the experiment were observed or analyzed in order to compare the dactylography, and confirm the source of the bacteria. RESULTS 1. Virulence experiment: among 20 mice of the experimental group, bacterial growing was found in the livers and lungs of 4 mice (20%). 2. Wound infection experiment: positive rate of the wound infection was 100%, the positive culture rate of the bacteria from the organs (of the sacrificed mice) was 30%, and in pathological test of the wounded legs, the following was found: inflammation in the subcutaneous tissue of the wounded legs, wound being infiltrated with a large number of neutrophils, muscle fiber being damaged, abscess being formed. CONCLUSIONS The V. alginolyticus derived from the sea water of the southeast coast area with concentration of 106 CFU/ml is strongly infective, it could lead to infect the experimental mice.
