A Mobile Model for Protein Adsorption Equilibrium by Electrostatic Interactions

摘要 根据统计热力学推导得到了反映蛋白质离子交换吸附非质量作用本质的可动平衡模型,并用阴离子交换剂吸附牛血清白蛋白的平衡数据进行了拟合分析.结果表明,可动吸附模型可定性分析离子强度及pH值对蛋白质吸附平衡的影响.在离子强度大于60mmol/L时,模型拟合精度较高;当溶液pH值较低并接近蛋白质等电点时,模型的拟合精度并未降低,从而克服了空间质量作用模型在高离子强度及pH接近等电点时预测效果差的缺点. A thermodynamic model—mobile model has been developed from the statistical thermodynamic theory for the nonlinear protein ion-exchange adsorption equilibrium.It gives a mechanistic description of protein adsorption in accordance with the non-mass action nature of protein ion-exchange adsorption.The adsorption layer nonidealities erising from protein-protein electrostatic and short-range interactions are considered.By comparison with batch ion-exchange adsorption equilibrium data of bovine serum albumin(BSA),the influences of pH and ionic strength on protein adsorption equilibrium are well interpreted by the model.In addition,comparison between the regression results obtained from the proposed model and steric mass-action (SMA) model is made.At ionic strengths higher than 60 mmol/L, the model agrees well with the equilibrium data.Moreover, the model accuracy is not declined even at the pH value close to the isoelectric point of BSA.This indicates the advantage of the model over SMA model.
