摘要
目的 观察负压治疗 (NP)对实验性缺血组织自由基损伤的保护作用。方法 采用切断犬股动脉分支 ,动脉腔内置入螺旋状金属丝的方法 ,在两周内复制出肢体动脉闭塞性病变的动物模型 ;对动物模型进行 8~ 10d的负压治疗后 ,采集模型建成及治疗前后的静脉血样 ,分别测定血中脂质过氧化物 (LPO)含量及超氧化物岐化酶 (SOD)活性并进行分析比较。结果 组织缺血后 ,LPO含量显著升高而SOD活性显著下降 ,二者与缺血前正常时相比较具有显著差异 [(35 .5 94± 10 .75 3)vs.(2 4 .80 0±9.0 11) μmol·L-1,P <0 .0 5 ;(10 3.90 0± 4 5 .82 1)vs.(15 7.2 5 2± 17.4 5 1)kU·L-1,P <0 .0 1];经 8~ 10d的负压治疗后 ,LPO含量明显下降而SOD活性显著升高 ,可恢复到模型建成前水平。对照组未经负压治疗 ,患肢静脉血中测得的LPO含量持续升高 ,SOD活性持续下降 ,与治疗组相比具有显著差异[(4 0 .0 0 0± 12 .5 4 3)vs.(2 4 .6 6 0± 4 .396 ) μmol·L-1,P <0 .0 1;(114 .4 94± 2 8.70 2 )vs.(16 6 .0 4 0± 34.72 1)kU·L-1,P<0 .0 5 ]。结论 肢体缺血后 ,局部组织的脂质过氧化反应增强 ,自由基清除能力下降 ,引起或加重组织损伤 ;肢体负压治疗可明显减轻脂质过氧化反应 ,增强自由基的清除能力 ,改善局部代谢状态 ,减?
Objective To observe the protective effects of le g negative pressure (NP) treatment on free radical injury from chronic ischemia of lower legs in canines. Methods By cutting off the branches of the femoral artery a nd inserting two pieces of spiral wires into the artery of canine posterior limb , we succeeded in duplicating the animal model of arterial occlusive disease in the lower extremity within two weeks. After treatment with LNP for 8 to 10 days, the blood samples of the ischemic legs were collected and the lipid peroxide (L PO) content as well as superoxide dismutase (SOD) activity were measured and ana lyzed, respectively. Results When the ischemic model was set up, PLO content inc reased while SOD activity decreased apparently, compared with those of normal [(35.594±10.753)vs. (24.800±9.011)μmol·L -1, P<0.05; (103.900±45.821)vs. (157.252±17.451)kU·L -1, P<0.01]. Aft er treatment with LNP for 8 to 10 days, PLO content declined and SOD activity in creased significantly. However, in the control group, this change was not seen. PLO content and SOD activity remained at a high level compared with NP-treated group [(40.000±12.543)μmol·L -1vs. (24.660±4.396)μmol·L -1, P<0.01; (114.494±28.702)vs. (166.040±34.721)kU·L -1, P<0.05]. Conclusion NP treatment can significantly diminish the pero xidation of ischemia and enhance the clearance ability of free radicals, which g ives rise to the severe damage to the tissues. It is concluded that NP has prote ctive effects on free radicals injury from chronic ischemia of lower legs.
