摘要 目的 建立筛选特异性芳香化酶抑制剂的筛选模型。方法 用胎盘的微粒体作为芳香化酶的酶源 ,以 [1β 3 H]雄烯二酮作为反应底物 ,通过测得产物3 H2 O的放射能来检测芳香化酶的活性。结果 建立了通过测定产物3 H2 O的放射能来检测芳香化酶活性的3 H2 O法。对该方法的反应动力学研究发现底物雄烯二酮的Vmax=33 3pmol·(min·mgpro tein) -1,Km=113nmol·L-1。用此方法作为芳香化酶抑制收稿日期 :2 0 0 4-0 3 -19,修回日期 :2 0 0 4-0 4-19资金项目 :国家重大科技专项“创新药物和中药现代化”资助项目作者简介 :全海天 ( 1980 -) ,男 ,博士生 ,研究方向 :肿瘤药理 ,Tel:0 2 1 5 0 80 660 0 2 5 17,E mail:haitianquan @hotmail.com ;楼丽广 ( 1967-) ,男 ,研究员 ,通讯作者 ,研究方向 :肿瘤药理 ,Tel:0 2 1 5 0 80 60 5 6,Fax :0 2 1 5 0 80 70 88,E mail:lglou @mail.shcnc .ac .cn剂的筛选模型 ,测得抗肿瘤药物exemestane对酶的抑制为不可逆抑制 ,其IC50 =70nmol·L-1,与文献报道相当。结论 此方法可以作为芳香化酶抑制剂的筛选模型 ,用来筛选以芳香化酶为靶点的药物。 Aim To develop an aromatase inhibitor screening a ssa y in vitro. Methods A method termed “tritiated water”assa y was developed to measure aromat aseactivity by quantifying the 3H 2O from the aromatization reaction, us ing placental microsomes as enzyme source and [1β- 3H]androstenedione as a substrate. Result “Tritiated water” assay was establishe d and it was found that androstrenedione had a K m value of 113 nmol·L -1 and a V max value of 33.3 pmol·(min·mg protein) -1 in the aromatiz ation reaction catalyzed by placental microsomes as determined by this assay. The reliability of t his assay was confirmed by application of specific aromatase inhibitor exemestan e. Exemestane was shown to be an irreversible aromatase inhibitor with the IC 50 of 70 nmol·L -1, which was comparable to findings by other laboratories.Conclusion These data demonstrate that “tritiated water” assay is a useful model to screen aromatase inhibitors.
