风火痰瘀同治对脑出血大鼠海马细胞色素C氧化酶活性与锥体细胞密度的影响 被引量:2

摘要 目的从神经元线粒体能量代谢角度探讨中医的风、火、痰、瘀同治法及其代表方———平肝熄风汤对大鼠脑出血神经元损伤的保护作用机制。方法采用胶原酶诱导大鼠脑出血模型 ,以组织化学方法结合灰度值半定量测定细胞色素C氧化酶 (cytochromecoxidase,CCO)活性 ,配合Nissl’s染色组织学观察锥体细胞密度和细胞计数。结果造模 1 2h,脑出血大鼠海马CA1区CCO活性较假手术组明显降低 (P <0 .0 1 ) ,随后呈进行性加重 ;2d出现锥体细胞明显缺失。用风、火、痰、瘀同治方药 (平肝熄风汤 )治疗后可维持其CCO活性 ,减少细胞缺失 ,维持锥体细胞密度。结论脑出血神经元损伤与神经元CCO活性降低有关。风、火、痰、瘀同治对脑出血继发性神经元损伤的保护机制可能与其能维持CCO活性、改善细胞有氧代谢、维持锥体细胞密度、保护线粒体功能有关。 ?Objective To investigate the protecting effects of treating method with wind, fire, phlegm and blood stasis resolved simultaneously on neurons in rats with cerebral hemorrhage and the mechanism of these effects responsible for mitochondrial energy metabolism.Methods The animal model with cerebral hemorrhage was prepared with the use of collagenase Ⅶ and the treating method studied here was taken to interfere with the modeling animals.Then, the activity of cytochrome C oxidase (CCO) was determined with the methods of histochemistry and semi-quantification of gray scals and the density of pyramidal cells was detected by cell counting on sections with Nissl's stain from hippocampi of the animals.Results Compared with that in sham group, the activity of CCO was significantly decreased in CA1 area of the rats of modeling group by 12 h following modeling (P<0.01)and it continuing to aggravate afterwards till 2d, when pyramidal cells showed significant loss.In contrast, the activity of CCO and the density of pyramidal cells were remained at the usual level in the animals treated with the studied treating method here (Liver-Calming and Wind-Extinguishing Decoction).Conclusions Neuronal injury of rats with cerebral hemorrhage is related to the decrease of CCO activity.The treating method with wind, fire, phlegm and blood stasis resolved together may bring about the neuronal protecting effects through maintaining CCO activity and pyramidal cell density at the usual level,improving aerobic metabolism and protecting mitochondrial function.
