摘要 目的 研究一氧化氮对胃粘膜的保护作用及其对胃粘膜保护因子的影响 ,探讨其胃粘膜保护机制。方法 雄性Wistar大鼠 80只 ,随机分为 4组进行 4项指标的检测 ,每组又分为 4个亚组 ,即对照组 L arg组 L NAME组 L NAME +L arg组 ,各组经不同药物处理后 ,检测 :①无水乙醇所致胃粘膜糜烂面积和深度 ;②通过Grees re action方法检测胃粘膜一氧化氮含量 ;③中性红分泌法及胃粘膜池模型方法检测胃粘膜血流 ;④血气分析检测胃粘膜中HCO3 -分泌。结果 L arg组大鼠给予无水乙醇灌胃后 ,其胃粘膜溃疡面积和深度显著低于对照组 ,L NAME组则显著高于对照组 (P <0 0 1) ,L NAME +L arg组胃粘膜溃疡面积和深度与对照组间差异无显著性意义。L arg可显著增加胃粘膜一氧化氮浓度 胃粘膜血流及胃粘膜HCO3 -分泌 ,给予L NAME后该促进作用消失 ,而单独给予L NAME可显著减低胃粘膜一氧化氮浓度 胃粘膜血流及胃粘膜HCO3 -分泌。结论 一氧化氮对胃粘膜有保护作用 ,其机制与增加胃粘膜血流和促进HCO3 -分泌有关。 Objective To observe the protective effect of nitric oxide on gastric mucosa and gastric mucosa protective factors, therefore, to investigate its protective mechanism. Methods Eighty male Wistar rats were randomly divided into 4 groups (n=20 in each) for detection of 4 parameters. Each group was assigned to the following subgroups: control subgroup, L-arg subgroup (L-arginine 200 mg/kg, i.p), L-NAME subgroup (L-NAME, 3 mg/kg, i.p) and L-NAME+L-arg subgroup. 1.5 ml 100 % alcohol was infused orally in all 4 groups. The area of gastric ulcer and depth was measured 15 min after the infusion. Levels of nitric oxide in gastric mucosa, blood flow of gastric mucosa and level of HCO_3-in gastric mucus were measured by Grees-reaction method, natural red secretion method and gastric mucosa chember method respectively. Results As compare with the control subgroups, the area and depth of gastric ulcer induced by alcohol were significantly decreased, and the level of nitric oxide, gastric mucosa blood flow and the level of HCO_3- were increased in L-arg subgroups. The protective effect was completely abolished by L-NAME. Whereas, L-NAME itself could increase the area and depth of gastric ulcer induced by alcohol, decrease the level of nitric oxide, gastric mucosa blood flow and level of HCO_ 3-. Conclusion Nitric oxide can prevent the gastric mucosa from the invasion factor and this effect is associated with the increased blood flow of gastric mucosa and secretion of HCO_3-.
