摘要
目的 建立测定小百部 (AsparagusfilicinusBuch . Ham)中小百部苷B含量的方法 ,评价不同产地药材的质量。方法 高效液相色谱法。色谱柱 :ZorbaxEclipseXDB C18柱 (5 μm ,15 0mm× 4 .6mm) ;流动相 :甲醇 水(82∶18) ;流速 :1.0mL/min ;柱温 :35℃ ;ELSD参数 ;漂移管温度为 10 0℃ ,气体流量 1.5L/min。结果 小百部苷B在 1.0~ 10 .0 μg范围内呈良好的线性关系 ,r =0 .9997,平均加样回收率为 96 .74 % ,RSD =1.90 % (n =5 )。结论 以HPLC ELSD法测定小百部中小百部苷B含量的方法可行。
Purpose: To establish a new the high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) method for the determination of aspafilioside B in the tuber of Asparagus filicinus. Methods The HPLC conditions were as follows: Zorbax Eclipse XDB-C18 column and the column temperature at 35°, drift tube temperature at 100°C with carrier gas flow being 1.5 L/min and water-methanol (82:18) as mobile phase with the flow rate being 1.0 mL/min. Results: There was a good linearity (r = 0.999 7) within the range of 1.0-10.0 μg. The average recovery of aspafilioside B was 96.74% with RSD of 1.90%. Conclusions HPLC-ELSD method was established and applied to the quantitative determination of aspafilioside B in the tuber of Asparagus filicinus, and aspafilioside B could be used as an indicator for quality assessment of Asparagus filicinus.
