摘要
本文对从玉米叶片提取的两种纯度(纯化和部分纯化)的PEP羧化酶在相对活性、km值、对G6P的敏感性和水分胁迫时酶活性的变化趋势等四个方面进行了比较研究,结果表明在以上四个方面用两个纯度PEP羧化酶所得结果几乎没有差异,因此可以认为在进行以上四方面研究时可以利用部分纯酶代替纯酶以简化程序和节省时间。同时本文对用粗酶代替纯酶用以测定酶活性的可能性进行了讨论。
In this thesis,the comparisons of relative activity,Km value,sensitivity to G6P and change of activity during water stress between purified and partly purified PEPCase from maize leaves were studied.There was little difference in relative activi- ty.Km value,sensitivity to G6P and tendency of activity change during water stress for two purities of PEPCase.We concluded that purified PEPCase could be replaced by part- ly purified PEPCase in research for relative activity,Km value,sensitivity to effector and change of activity during environmental stress.The possibility that crude PEPCase re- placed purified PEPCase and used as the source of PEPCase for measuring activity was also discussed.
