摘要
目的:探讨针灸对去卵巢大鼠骨质疏松症的作用机制和环节。方法:骨组织TGF-β_1 mRNA、VEGF mRNA采用原位杂交,凋亡基因Fas采用免疫组织化学检测。结果:模型组TGF-β_1、VEGF光度低于假手术组(P<0.05),针刺组、艾灸组、雌二醇组TGF-β_1、VEGF光度高于模型组(P<0.05);骨组织FAS蛋白光度,模型组高于假手术组(P<0.01),针刺组、艾灸组和雌二醇组低于模型组(P<0.05~0.01);成骨细胞内FAS蛋白阳性表达率模型组高于假手术组(P<0.01),雌二醇组低于模型组(P<0.01);破骨细胞内FAS蛋白阳性表达率针刺组、艾灸组和雌二醇组高于假手术组(P<0.05~0.01)。结论:针灸能有效调节破骨细胞内Fas基因的表达;能激活TGF-β_1、VEGF活性,使TGF-β_1 mRNA、VEGF mRNA表达增加。
Objective: To explore the mechanism of acupuncture and moxibustion for treatment of osteoporosis in ovariectomized rats. Methods: TGF-β1, mRNA. VEGF mRNA expression of bone tissue and apoptosis gene Fas were detected respectively by in situ hybridization and immunohistochemistry. Results: TGF-β1 and VEGF gray density in the model group were lower than those in the sham operation group (P<0. 05), and those in the acupuncture group, the moxibustion group and the estradiol group were higher than those in the model control group (P< 0. 05). Fas protein positive expression rate in osteoblast in the model group was ligher than that in the sham operation group (P<0. 01). and in the estradiol group was lower than that in the model group (P<0. 01). Fas protein positive expression rate in the osteoclast in the acupuncture group, moxibustion group and estradiol group were higher than those in the sham operation group (P < 0.05 - 0.01). Conclusion: Acupuncture and moxibustion can effectively regulate Fas gene expression in osteoblast. stimulate activities of TGF-β1 and VEGF, and increase expression of TGF-β1, mRNA and VEGF mRNA.
