摘要
目的观察大黄总蒽醌对SD大鼠所发生的各种毒性反应 ,以评价大黄总蒽醌的安全性。方法大黄总蒽醌以 0、14 0、794、4 5 0 0mg/kg,ig ,1次 /d ,每周 6次 ,连续给药 2 6周。结果ig高剂量后 ,大鼠精神不佳 ,排稀便、软便 ,体重增长缓慢。在给药 11周时 ,高剂量组 1只大鼠濒临死亡。高剂量组红细胞计数、血红蛋白含量、红细胞压积和Na+ 显著低于对照组 ,而尿素氮、总胆固醇、尿酸、K+ 和Ca2 + 升高 ,尿 β 微球蛋白、总蛋白质等也显著升高。高剂量组肾脏均可见近曲小管上皮细胞不同程度肿胀变性。结论在该试验条件下 ,大黄总蒽醌对SD大鼠的毒性反应靶器官可能主要是肾脏 (特别是肾近曲小管 ) ,这种毒性反应是可逆的 ,对SD大鼠的安全剂量为 794mg/kg。
PurposeThe toxicity study of total anthraquinone in Radix et Rhizoma Rhei, was conducted in SD rats to evaluate its safety.MethodsTotal anthraquinone in Radix et Rhizoma Rhei was administered to rats at oral doses of 0, 140, 794, 4 500 mg/kg once daily for 6 months.ResultsIn the group treated with 4 500 mg/kg, a major toxic sign was cachexia, diarrhea, and body weight increasing slowly. On week 11th of administration, 1 rat was closed to death in the high dose group. Hematological and blood chemical tests revealed a decrease in RBC, HGB, HCT and Na+, and notabe increase in UA, BUN, TC, Ca 2+ and K+ was noted in the 4 500 mg/kg dose group. Urinalysis test results showed thal urine β 2-MG, Alb, Glu, etc. were notably increased. Histopathological examination showed tumefaction and degeneration in renal proximal tubules epithelia in all animals from high dose group sacrificed on week 13 and 26.ConclusionFrom these results, the safe-dose of Rhubarb total anthraquinones, was estimated to be 794 mg/kg under the present conditions, and the toxic target organ was estimated to be kidney (esp. the proximal tubules).
