摘要
目的 观察碳酸缓冲液干预对光气染毒后大鼠AT-Ⅱ细胞基因表达的影响,以深入探讨其作用的分子机制。方法分别抽提正常组、损伤组及干预组AT-Ⅱ细胞总RNA,通过逆转录方法,进行荧光标记后,制备成cDNA探针,与表达谱芯片进行杂交及扫描测定,通过计算机进行数据处理、基因库对比并筛选出差异表达基因。结果 经碳酸缓冲液干预后,筛选出的差异表达基因数由单纯损伤组的161条降低到了115条,且大部分与肺损伤相关基因的异常表达均得到了抑制,并出现了新的差异基因。结论碳酸缓冲液在干预光气中毒中具有一定的保护作用,为探讨光气中毒和临床治疗机制提供了新的思路。
Objective To study the molecular mechanism of rat acute lung injury caused by phosgene with the treatment of carbonic acid buffer and to screen differential genes expressions. Methods Rats were divided into control, injury and treated groups. The original generation cells of rat AT- E from each group were cultured and the total RNAs were isolated from AT- Ⅱ cells. Then the total RNAs were reversely transcribed to cDNA with the incorporations of fluorescent Cy3 ., Cy5, for preparing the hybridization probes. The mixed probes were then hybridized to gene chip. The gene chip were scanned for the fluorescent signals and the differential genes were analyzed with software. Results After treating with carbonic acid buffer, the number of differential genes decreased from 161 in the injury group to 115 in the treated group. The majority of all differential genes related to lung injury were inhibited but some new differential genes arised. Conclusion Carbonic acid buffer played some protection effect on phosgene injury, which may provide some new thought for clinical therapy to phosgene injury.
