摘要
以自制桦木木聚糖为原料,采用稀硫酸水解,活性炭柱分离,冷冻干燥结晶的方式制备低聚木糖单一组分标准品。稀硫酸浓度为0.25mol/L,水解温度100℃,水解时间15min。水解液上活性炭柱流速50mL/h,洗脱液为30%乙醇,梯度淋洗,洗脱速率100mL/h。冷冻干燥条件-50℃,真空度5.33Pa。酸解产物以木二糖、木三糖、木四糖、木五糖、木六糖为主。
This paper described a method for preparing and isolating xylooligosacc harides with three steps: hydrolyzing the birch xylan with sulfuric acid solutio n, separating the hydrolysis solution with an activated charcoal column and gett ing single xylooligosaccharide by freeze drying. The concentration of the sulfur ic acid was 0.25mol/L.The hydrolysis temperature was 100℃ and the reaction time was 15min. The hydrolysis solution flow rate was 50mL/h. The single xylooligosa ccharide absorbed on the activated charcoal was eluded with 30% alcohol. Each xy looligosaccharide was crystallized by freeze drying at-50℃.The resulted produc ts were composed of disaccharide,trisaccharide,tetrasaccharide,pentasaccharide, hexasaccharide.
