摘要
采用分割切块法测定了 3个草菇菌株 (V0 1、V0 2和V0 3)在低温下的致死时间。结果表明 :V0 1、V0 2和V0 3菌株在 0、 2和 4℃下的致死时间分别为 2 4、 4 8和 32h ,4 8、 88和 6 4h ,96、 192和 14 4h。用 0 0 1%吖啶橙诱变草菇菌株V0 1担孢子 30min ,致死率达 70 4 % ;对诱变处理的担孢子萌发形成的菌体进行低温临界致死处理 ,杀死低温敏感菌株 ,采用覆层平板法筛选出
The critical cryolethal time of three Volvariella volvacea strains V01,V02,V03 under 0 ℃,2 ℃ and 4 ℃ were found to be 24 h,48 h,32 h;48 h,88 h,64 h;as well as 96 h,192 h,144 h,respectively.The basidiospores of V01 were treated with 0 01% acridine orange at 25 ℃ for 30 min and the killing ratio reached to 70 4%.After spore germinating on IPDA plates,the colonies were treated for the critical cryolethal time to kill the sensitive ones.With cladding plate method,four low temperature resistent strains were selected out.
