摘要
目的研究大鼠全脑缺血再灌注损伤后海马c fos表达和神经元的凋亡。方法wistar大鼠双侧颈总动脉夹闭 30min ,造成全脑缺血 ,松开动脉夹再灌注。动物均于再灌注 1h行c fos原位杂交 ,再灌注 4 8h行TUNEL染色 ,观察海马神经元的凋亡情况。结果缺血再灌注后 ,大鼠海马CA1区c fos基因表达增强 ,缺血再灌注大鼠的TUNEL染色可见CA1区有大量阳性细胞 ,与对照组相比有显著性差异 (P <0 .0 1)。结论脑缺血再灌注后导致海马c fos基因的表达显著增强 ,细胞凋亡明显增加。
Objective To investigate c-fos expression and neuron apoptosis after cerebral ischemia-reperfusion damage in rats.Methods An ischemia model of wistar rat was established by clipping bilateral cervical arteries for 30 minutes, and then reperfusion was made through removing the clipping. Then,the gene expression of c-fos was detected 1 hour and TUNEL staiming was made 48 hours after reperfusion.Results The gene expression of c-fos exhibited a significant increase and many positive cells were observed with TUNEL assay following cerebral ischemia-reperfusion damage in comparison with normal rat(P<0.01).Conclusion The cerebral ischemia-reperfution can enhance c-fos expression and neuronal apoptpsis.
