黄芪多糖和氧化苦参碱协同增强鸡新城疫疫苗免疫效果研究 被引量:22

摘要 【目的】研究黄芪多糖(Astragalus polysaccharides,APS)和氧化苦参碱(Oxymatrine,OM)对新城疫疫苗免疫的影响。【方法】将14 日龄非免疫罗曼蛋公鸡550 羽随机均分为11 组,1~9组分别为OAP(以OM与APS质量比为6∶4配成)、OM和APS的高(4 mg/mL)、中(2 mg/mL)、低(1 mg/mL)剂量组,10组为免疫对照组,11组为空白对照组。除空白对照组外,各组试验鸡只均于14日龄首免NDVⅣ系疫苗,28日龄重复免疫。在首次免疫的同时,1~9 组分别口服高、中、低剂量药物,免疫对照组和空白对照组口服等量生理盐水,每天1 次,连续3 d。分别于14,21,28,35和42 日龄采血,用MTT法测定T淋巴细胞增殖的变化,用β微量法测定血清抗体效价变化。于14,21,28和35 日龄,每组随机抽取鸡4 羽,称体质量,剖杀,摘取法氏囊和脾脏并称质量,计算免疫器官指数。【结果】OAP、OM、APS高、中剂量在各时间点均能促进T淋巴细胞增殖;OAP高、中剂量组在各时间点抗体效价显著高于免疫对照组;OAP高、中、低剂量组在各时间点的脾指数和法氏囊指数均显著高于免疫对照组(OAP低剂量组35日龄除外)。【结论】黄芪多糖和氧化苦参碱能协同增强鸡新城疫疫苗的免疫效果。 【Objective】This paper aimed to study the immune effect of astragalus polysaccharide(APS)and oxymatrine(OM)on Newcastle disease vaccine.【Method】Five hundred and fifty 14-day-old non-immunized Roman cocks were randomly divided into 11 groups.Groups 1to 9 were OAP(OM and APS quality ratio of 6∶4),OM and APS at high(4mg/mL),medium(2mg/mL)and low(1mg/mL)doses,respectively.Group 10 was vaccine control and group 11 was blank control.Groups 1to 10 were vaccinated with NDV-Ⅳat the ages of 14 and 28days,respectively.At the same time of the first vaccination,the chickens in groups 1to 9were given compounds at high,medium and low doses and chickens in vaccination and blank control groups were given physiological saline by oral administration,once a day for three days.On days 14,21,28,35 and 42,peripheral T lymphocyte proliferations and serum antibody titers were deter-mined with MTT method andβ-microscopic method,respectively.On 14,21,28 and 35days,the organ indexes of spleen and bursa were determined by killing 4chickens in each group.【Result】T lymphocyte proliferations were promoted by OAP,OM and APS at high and medium doses and serum antibody titers of OAP at high and medium doses were significantly higher than those of vaccine control group.At any time,spleen and bursa indexes of OAP at all doses were significantly higher than those of vaccine control group(except the OAP low dose group for 35-day old chicken).【Conclusion】APS and OM could synergistically improve the immune efficacy of Newcastle disease vaccine in chicken.
