应用DIBA法检测侵染辣椒的TMV与CMV的研究

摘要 采用斑点免疫结合法(Dot Immunobinding Assay,DIBA)检测侵染辣椒的TMV与CMV均具有极强的专化性.在可检测的感染叶片的汁液稀释范围内,用TMV与CMV的抗血清分别交叉检测CMV与TMV,均未出现阳性反应.检测感染叶片粗汁液的最高稀释度TMV为1:5 120;CMV为1:2560.利用3种不同封闭系统[TBST,TBST+3%牛血清白蛋白(3%BAs),TBST+3%酪蛋白(3%Casein)]检测,比较结果表明;采用TBST+3%BAS与TBST+3%Casein封闭效果较好:与免疫电镜(ISEM)检测TMV与CMV相比较,DIBA敏感性分别高出15与7倍. Dot Immunobinding Assay (DIBA) is a very specific method in the detection of TMV and CMV in pepper plants. There is no positive reaction in the detections of CMV with TMV-Antiserum and of TMV with AMV-Antiserum respectively. The maximum reacting dilutions of crude sap (TMV) are 1:5 120 and CMV are 1:2 560.The coparison of three blocking systems (TBST, TBST+3%BSA, TBST+3%Casein) in TMV and CMV detection showed that both TBST+3%BSA and TBST+3% Casein are better than TBST. The sensitivity of detection of sap extracts of infected pepper leaf with TMV or CMV by DIBA is 15 times and 7 times more than that by Immuno Sorbant Electron Microscopy (ISEM).
