摘要
弧菌是危害水产养殖动物的最为严重的病原之一。从患病大黄鱼肝脏分离致病的细菌哈维氏弧菌GYC1108-1为材料,对哈维氏弧菌GYC1108-1株的最佳生长条件及培养基优化进行了测定。综合考察不同的培养温度、培养时间、培养基盐度、培养基的pH值对细菌生长的影响,并采用正交实验法,对培养基配方进行优化。结果表明:不同的培养温度、培养时间、培养基盐度、培养基pH值等均会影响细菌的产量。GYC1108-1适宜生长的盐度为1%~5%、pH为5~9.5、温度为15~35℃、在培养基中添加硫酸铜、硫酸氨会明显促进生长;最佳培养条件及培养基成份为:NaCl 2%,pH8.0,温度30℃,蛋白胨1.0%、牛肉膏0.75%、CuSO_4 0.3mg/L、(NH_4)_2SO_4 0.1g/L。
Vibrio harveyi was isolated from the diseased Pseuclosciaena crocea's live.And the optimal condition was studied. Meanwhile,a set of experiments designed by the orthogonal method was adopted to optimize the culture medium elements.It showed that all the different conditions can affect the growth of GYC1108-1.The suitable salinity of Vibrio harveyi was 1%to 5%,pH was 5 to 9.5,temperature was 15℃to 35℃,CuSO4 and(NH4)2SO4 in the culture medium can promote the growth of bacteria.The optimal growing condition of Vibrio harveyi was concluded:salinity 2%,pH 8,and temperature 30℃.In addition,the optimized elements of culture are:peptone 1.0%,beef extract 0.75%,CuSO_4 0.3mg/L,(NH_4)_2SO_4 0.1g/L.
