摘要
目的观察狼疮清颗粒对主动型Heymann肾炎(AHN)模型大鼠肾功能的作用及其对MRL/lpr小鼠肾组织NF-κBp65蛋白表达的影响。方法大鼠肾皮质加弗氏完全佐剂腹腔注射建立AHN大鼠模型,应用狼疮清颗粒、泼尼松、狼疮清颗粒联合泼尼松,对AHN模型大鼠进行干预治疗4 w,检测其24 h尿蛋白、尿素氮(BUN)、肌酐(Cr)及肾脏病理学变化,同时对MRL/lpr小鼠干预治疗4 w,Western印迹检测肾组织NF-κBp65蛋白表达。结果与模型组相比,各治疗组均降低AHN模型大鼠尿蛋白、BUN、Cr的水平,不同程度的改善AHN模型大鼠肾组织病理变化;显著降低MRL/lpr小鼠NF-κBp65蛋白的表达,其中以狼疮清颗粒联合泼尼松组(中西医结合治疗组)最为明显。结论狼疮清颗粒对狼疮性肾炎具有一定的治疗作用,其机制可能与抑制肾组织NF-κBp65蛋白表达有关。
Objective To investigate the effects of langchuangqing granule on renal function in rats of active heymann nephritis and the expression of NF-κBp65 protein on renal tissue in the MRL / lpr mice.Methods AHN rats were produced by intraperitoneal injection of renal cortex and Freund's complete adjuvan.Intervention therapy with langchuangqing granule,prednisone,langchuangqing granule + pred- nisone were applied respectively to AHN rats for 4 weeks.24 h urine protein,urea nitrogen,creatinine and pathological changes were meas- ured.Intervention therapy was applied to MRL / lpr mice for 4 weeks in order to detect the expression of protein NF-κBp65 in renal tissue by Western blot method.Results To compare treatment groups with the model group,it was revealed that levels of urine protein,urea nitrogen and creatinine in AHN rats and the expression of protein NF-κBp65 in renal tissue in MRL / lpr mice were significantly decreased.Pathologi- cal changes in renal tissue in AHN rats were improved at different degree also.Effect in the treatment group which integrated traditional Chi- nese and western medicine(langchuangqing granule + prednisone) was most significantly.Conclusions Langchuangqing granule has cer- tain therapeutic effect on lupus nephritis.The mechanism is thought to be related to the decreased expression of protein NF-κBp65.
