透骨消痛胶囊在大鼠体内药代动力学初步研究 被引量:3

摘要 目的以透骨消痛胶囊为例,探讨吸光度-波长曲线下面积应用于中药复方药代动力学研究的新思路与方法。方法通过配制系列浓度的透骨消痛胶囊内容物的溶液,在200~400 nm下进行全波长扫描,观察吸波面积和总药物浓度的线性关系,并对透骨消痛胶囊开展灌胃后大鼠体内药代动力学研究。结果不同浓度内容物溶液的吸波面积和内容物中药物的总浓度具有良好的线性关系,归一化方程为:AUAWC=0.6608x-0.1206,(r=0.9999)。大鼠体内透骨消痛胶囊内容物灌胃后,Ka=7.72/h,t1/2(Ka)=0.0898 h,吸收分布较迅速;T(peak)=0.4656 h,第0.5 h左右达到最大血药浓度,Cmax=33.9576μg/mL;第6 h出现双峰现象,说明可能存在肠肝循环;Ke=0.2368/h,t1/2(Ke)=2.9271 h,药物消除较慢,总浓度维持较长的时间。结论吸波面积和中药总浓度成线性的关系可应用于多成分复杂体系的中药药代动力学研究,不同时间段获得的中药总浓度能较好地体现复杂成分的总量在体内的'动态'过程。 Objective To explore a new idea and method of area under absorbance-wavelength curve(AUAWC) used in the pharmacokinetic study of traditional Chinese medicine(TCM) with Tougu Xiaotong capsule as an example. Methods By preparing a series of solutions with different concentrations of Tougu Xiaotong capsule, the linear relationship between AUAWC and total drug concentration was observed after a full ultraviolet-visible spectrum scan to samples from 200 to 400 nm wavelength. The pharmacokinetic study in rats was carried out after the rats received Tougu Xiaotong capsule by gastric perfusion. Results A good linear relationship between AUAWC of solutions with different concentrations and total drug concentration was existed, and the normalized equation was AUAWC=0.6608x-0.1206(r=0.9999). When the rats were administrated with certain concentration of Tougu Xiaotong capsule, Ka was 7.72 / h and t1 / 2(Ka) was 0.0898 h, which showed the absorption and distribution were rapid; Tmax was 0.4656 h and Cmax was 33.9576 μg / mL, and it reached the maximum drug concentration in the blood after about 0.5 h; a bimodal phenomenon appeared after 6 h, which indicated enterohepatic circulation could be existed; Ke was 0.2368 / h and t1 / 2(Ke) was 2.9271 h, and the drug elimination was slow and the total concentration was maintained in a long time. Conclusion The linear relationship between AUAWC and total drug concentration could be well applied to pharmacokinetic study of TCM with multi-components complex system. The total concentration of TCM obtained in different periods could better reflect the 'dynamic' process of the total amount of complex components in rats.
