摘要
目的观察麦冬提取液对人肺癌细胞株SPC-A1细胞放射效应的影响。方法实验分6组,空白对照组、单纯照射组、单纯药物组、照射前给药组、照射中给药组和照射后给药组。应用集落形成实验测定各组细胞的存活率,计算麦冬提取液的放射增敏比;流式细胞法分析比较各组细胞周期的变化。结果在2Gy照射剂量下,麦冬提取液与照射联合应用组的存活率低于单纯照射组,并以照射后加药组相对更为明显(P<0.05);在放射前、中、后联用麦冬提取液的各组中,放射增敏比分别为1.11、1.46、1.29。流式细胞法分析显示,麦冬提取液与照射联合应用组G2/M期比例上升,与空白对照组、单纯照射组、单纯药物组比较均具有统计学意义(P<0.05)。结论麦冬提取液对经照射的人肺癌细胞株SPC-A1细胞具有一定的增效作用,其机制可能与改变细胞周期有关。
Objective To observe the effects of extracting solution of Ophiopogonis Japonicus(ESOJ) on radiochemy of human lung cancer cell lines SPC-A1. Methods The cells were randomly divided into control group, irradiation group, ESOJ group, group received ESOJ before irradiation, group received ESOJ during irradiation, group received ESOJ after irradiation. The survival fraction of cells in each group was de-tected respectively by colony formation assay, the radiosensitization ratio of ESOJ was calculated, and the changes of cell cycle among the six groups were compared by flow cytometric analysis. Results When the dose was increased to 2 Gy, the survival fraction in irradiation combined with ESOJ groups was respectively lower than that of irradiation group, and the difference was significant in group received ESOJ after irradiation(P<0.05). The radiosensitization ratio was 1.11, 1.46, 1.29 in group received ESOJ before irradiation, group received ESOJ during irradiation, group received ESOJ after irradiation respectively. Flow cytometric analysis showed the ratio in G2 / M phase was increased significantly compared with control group, irradiation group and ESOJ group(P<0.05). Conclusion ESOJ has a certain synergistic action on the radiochemy of human lung cancer cell lines SPC-A1 received irradiation, and the mechanism may be related to the change of cell cycle.
