哮喘患者外周血Rho激酶及CD4^+ CD25^+调节性T细胞测定 被引量:8

Measurement of Rho-kinase and CD4^+CD25^+ regulatory T cells in the peripheral blood in asthmatic patients

摘要 目的:比较不同研究对象外周血Rho激酶及CD4+CD25+调节性T细胞的表达规律,探讨Rho激酶和CD4+CD25+调节性T细胞在哮喘患者中的变化和相关性。方法:选取中、重度哮喘患者16例,健康对照组14例,通过年龄、性别配对,ELISA测定所有研究对象血清Rho激酶水平,流式细胞仪测定CD4+CD25+调节性T细胞水平,所有研究对象经肺功能仪测定肺功能。结果:哮喘组外周血Rho激酶表达水平高于对照组(P<0.05),外周血CD4+CD25+调节性T细胞水平低于对照组(P<0.05);两组外周血Rho激酶与肺功能第1秒用力呼气容积(forced expiratory volume in one second,FEV1)占预计值百分比(FEV1%)无相关关系(r=-0.491,P>0.05);两组外周血CD4+CD25+调节性T细胞水平与肺功能FEV1%呈轻度正相关关系(r=0.380,P=0.038);两组外周血Rho激酶与CD4+CD25+调节性T细胞水平无相关关系(r=-0.438,P>0.05)。结论:哮喘患者外周血Rho激酶水平升高,CD4+CD25+调节性T细胞水平降低,可能在哮喘发病过程中起一定的作用。 Objective: To determine the levels of Rho-kinase and CD4+CD25+ regulatory T cells in patients with asthma, and the relationship between Rho-kinase and CD4+CD25+ regulatory T cells.Methods: We included 16 patients with moderate to severe asthma in the research group and 14 healthy people as the control group. The levels of Rho-kinase in the 2 groups were measured by ELISA. The level of CD4+CD25+ regulatory T cells in the 2 groups was measured by flow cytometry. The pulmonary function was measured by spirometer.Results: The level of Rho-kinase in the research group was higher than that in the healthy controls(P<0.05). The level of CD4+CD25+ regulatory T cells in the healthy controls was higher than that of the research group(P<0.05). There was no correlation between the level of Rho-kinase in the peripheral blood of the 2 groups and forced expiratiory volume at the first second/ forced vital capacity(FEV1%)(r=-0.491, P>0.05). The level of CD4+CD25+ regulatory T cells in the peripheral blood of the 2 groups showed a positive correlation with FEV1%(r=0.380, P=0.038). There was no correlation between the level of Rho-kinase and the level of CD4+CD25+ regulatory T cells in the peripheral blood of the 2 groups(r=-0.438, P>0.05).Conclusion: Rho-kinase and CD4+CD25+ regulatory T cells may play a key role in the pathogenesis of asthma.
