微柱离心-高效液相色谱法测定莪术醇脂质体的包封率 被引量:7

摘要 目的建立莪术醇脂质体包封率测定的微柱离心-高效液相色谱法。方法采用葡聚糖凝胶(Sephadex G-50)微柱离心法分离脂质体和游离药物,采用HPLC法测定脂质体中药物含量,计算包封率。结果葡聚糖凝胶G-50对空白脂质体无吸附,最佳离心条件为3000r·min-1、2min,含量测定的线性范围为2.0~16.0mg·L-1,高、中、低3个质量浓度的日内、日间RSD均小于2%,回收率为99.82%~101.61%,3批样品中莪术醇的平均包封率为73.62%。结论微柱离心-高效液相色谱法可作为莪术醇脂质体质量控制的有效手段之一。 Objective To establish a minicolumn centrifugation-high performance liquid chromatography method for determination of entrapment efficiency of curcumol liposomes.Methods The liposomes were separated from free curcumol using Sephadex G-50-filling minicolumn by centrifugation.High performance liquid chromatography was developed to determine the entrapment efficiency of curcumol in liposomes.Results By flowing through the Sephadex G-50 minicolumn,blank liposomes were not absorbed.The condition of centrifugation was 3 000 r·min-1for 2 min.The calibration curve was linear in the range of 2.0-16.0 mg·L-1 for curcumol.The precision of inter-day and intra-day was less than 2 %,and the recoveries of the method were in the range of 99.82 %-101.61 %.Conclusion The method can be used for the quality control of curcumol liposomal formulation.
