响应面法优化烟草游离烟碱的提取工艺研究 被引量:2

Optimization of Unprotonated Nicotine Extraction Technology from Tobacco by Response Surface Method

摘要 为研究提取因素对烟草中游离烟碱的提取得率的影响。在单因素试验基础上,选择提取溶剂中液料比、提取时间、提取温度和摇床转速,进行4因素3水平Box-Behnken试验设计,采用响应面法(RSM)分析了4个因素对响应值(游离烟碱得率)的影响。结果表明,水提取-有机溶剂萃取法提取烟草中游离烟碱的最优条件如下:液料比30∶1(V/m,mL/g),提取时间33 min,摇床转速212 r/min和提取温度34℃。在最优的条件下进行了5次验证试验,游离烟碱的平均得率为7.01 mg/g,与理论值(6.98 mg/g)的误差为0.43%,说明该优化方法可行。 Response surface analysis methodology(RSM) was applied to optimize the conditions for extracting unprotonated nicotine from tobacco. Based on single factor experiments, four-factors-three-levels Box-Behnken central composition experiments design were applied. The ratio of liquid to solid, extraction time, extraction temperature and rotating speed of table concentrator were chosen as effect factors. The results showed that the optimum conditions were ratio of liquid to solid, 30∶1;extracting time, 33 min; extracting temperature, 34 ℃; rotating speed of table concentrator, 212 r / min. 5 verification experiments at these optimum conditions showed an average yield of uprotonated nicotine at 7.01 mg / g, while the predicted yield was 6.98 mg / g; the RSD was 0.43%, indicating that this method was feasible.
