全反式维甲酸对糖尿病心肌病大鼠心功能的影响 被引量:1

The Influence of All-trans Retinoic Acid on Cardiac Function of Rats with Diabetic Cardiomyopathy

摘要 目的:探讨全反式维甲酸(ATRA)对糖尿病心肌病(DCM)大鼠心功能的影响。方法:20只Wistar大鼠腹腔注射链脲佐菌素制作糖尿病模型,4周后随机取10只予ATRA20 mg/(Kg.d)灌胃(维甲酸组),另10只予等量纯净水(糖尿病组)。8周后分别测量各组左室重量指数(LVMI),左室收缩压(LVSP)、左室舒张末期压(LV-EDP)、室内压最大上升速率(+dp/dtmax)、室内压最大下降速率(-dp/dtmax)和室内压上升达最大速率所需时间(t-dp/dt),电镜观察心肌细胞超微结构。另设对照组。结果:全反式维甲酸可使糖尿病大鼠LVMI、LVEDP和(t-dp/dt)值降低、LVSP、(+dp/dtmax)和(-dp/dtmax)值升高,其可明显减少心肌间质内胶原堆积和线粒体肿胀,肌原纤维断裂、溶解现象较轻,细胞排列较整齐;微血管腔内皮细胞肿胀较糖尿病组轻。结论:全反式维甲酸可改善糖尿病大鼠心脏收缩和舒张功能,减轻心肌肥厚,对糖尿病大鼠心肌有保护作用。 Ob je c tive To investigate the influence of all trans retinoic ac id(ATRA) on card iac func tion of rats with diabetic ca rdiomyopa thy. Me thods The d iabetic mode l were established in 20 ra ts by intraper itoneal injection of strep tozotoc in( 20 mg/kg). Then the ra ts were random ly d ivided intoATRA group and DCM group after 4 weeks. The ra ts in ATRA group were given ATRA w ith 20 mg/( kg d) by intragastric adm in istration, while equal volume purified wate r were given in DCM group. The le ft ventr icularmass index( LVMI), le ft ventr icular systolic pressure(LVSP) , left ventr icu lar end diastolic pres sure( LVEDP), the maxima l rates of increase /dec line of left ventricu lar pressure ( dp /dtmax ), the maxima l rates of de c rease of le ft ventr icular pressure ( - dp/ dtmax ) and the time of the maximal ra tes of decrease of left ventr icular pressure ( - dp /dtma x ) were exam inated at 8 weeks la ter. The myocardia l ultrastruc turewere obse rved w ith e lectronm ic roscope. Re s u lts A ll trans re tinoic ac id cou ld decreasethe values of LVM I, LVEDP and ( t dp /dt), wh ile increase the LVSP, ( + dp / dtmax ) and ( - dp/ dtmax ) of rats with diabetic.
