摘要   基因治疗中导入基因表达时间和水平的调控极其重要.哺乳动物细胞中用到的诱导型载体主要与启动子有关,如热休克蛋白(Hsp)启动子可在高温下被诱导,还有重金属、糖皮质激素诱导的启动子等,但这些系统均存在着诱导表达不具备特异性、系统处于关闭状态时表达有遗漏以及诱导剂本身具有毒性会对细胞造成损伤等缺陷.如果设置一种导入基因的'开关',能调控基因的表达,则有望大幅度提高基因治疗在临床应用的安全性.1992年,Gossen和Bujard[1]首次成功地利用原核基因调控元件构建了真核细胞基因表达调控系统--四环素基因表达调控系统(Tet系统),该系统能通过在培养基中加入或者去除四环素或其衍生物(如强力霉素)诱导或抑制所感兴趣基因的表达,故被广泛应用于基因表达调控、蛋白质功能和基因功能研究以及基因治疗研究中.本文就近年来Tet系统的研究进展等做一综述.……
