摘要
为获得高质量的鸭肝组织RNA样本,通过优化提取方案,从1 g超低温(-80℃)的冻存组织中提取RNA,以紫外分光光度计对RNA的含量进行了测定,并对其纯度进行了分析;通过琼脂糖凝胶电泳对RNA的完整性进行了检测,并对所得样本的稳定性进行了测试。结果表明:试验得到了1 528μg RNA,样品OD260/OD280=1.93,电泳得到的28 s和18 s RNA条带清晰可见,没有发生降解,样品在室温条件可保存24 h以上,在-20℃可保存8 d以上。提取的RNA质量较高。
In order to obtain high quality total RNA from duck liver tissues stored at-80℃,the RNA was extracted by optimizing the extraction scheme,the RNA content and purity was assayed by ultraviolet spectrophotometer,the integrity was detected by agarose gel electrophoresis,and the stability of the extracted RNA was tested in the study.The result showed that 1528 μg RNA was extracted from 1 g liver tissue,OD260/OD280=1.93 in the sample,the 28s and 18s RNA were obviously absolved,the RNA sample did not degraded and could be stored more than 24 hours at room temperature,more than 8 days at-20℃,the activity of sample could effectively support future study.The study indicated that the quality total RNA extracted from the duck liver tissues was higher.
