摘要
通过滤纸床发芽方法测定一串红种子发芽率。采用 HPLC测定种子发育及贮藏过程中内源 GA3和 ABA含量 ,并观察外源 GA3、ABA以及去掉种皮对种子休眠及发芽的影响。结果表明 ,新采收的不同成熟度的种子发芽率不同 ,未完全成熟的二级种子最高 ,完全成熟的一级种子次之 ;随成熟度提高 ,种子中内源 GA3含量逐渐下降。在室温或低温干燥贮藏条件下 ,随贮藏时间延长 ,发芽率增加 ,3个月左右达到最大值 ;干种子内源 GA3含量却随贮藏时间延长而降低 ,仅在后期有所上升 ;外源 GA3处理对新采收和贮藏 1 -2个月的种子发芽率促进作用明显 ,但随贮藏时间延长而减弱。去掉种皮 ,裸胚发芽率增加幅度很大。在种子发育及贮藏过程中均没测出 ABA,外源 ABA处理种子 。
In this experiment, filter paper bed method was used to measure the germinating rate of Salvia splendens. The endogenous GA 3 and ABA contents were detected by HPLC. The results indicated that the seed germinating rates were different, depending on maturity degrees of the seeds just after harvesting. The second grade seeds fully unmatured had the highest germinating rate, and the first grade seeds fully matured were following. Endogenous GA 3 content decreased gradually in the seeds with the seed maturity. The seed germinating rates were increased with extension of storage time, being the highest rate for 3 months after storage, whatever the seeds were stored under room condition or under the dried low temperature condition. However, the endogenous GA 3 content decreased followed by extension of the storage time and had a slightly increase only by the end of storage. Application of GA 3 had a remarkable effect on the seed germinating rate only for seeds of both new harvested and stored for 1 to 2 months. The germinating rate was highly increased by removing the seed coat. No ABA contents were detected in the seeds either in the seed development or during the seed storage, and the seed germinating rates had no remarkable changes by applications of ABA.
