酵母双杂交技术筛选肺癌组织中MCM7蛋白结合蛋白 被引量:3

摘要 背景与目的微小染色体维持蛋白(mini-chromosome maintenance proteins,MCM)MCM7为MCM复合体成员之一,在启动DNA复制、合成中起关键作用,但对能够与其结合的蛋白所知甚少。本研究应用酵母双杂交方法筛选肺癌组织中MCM7蛋白的结合蛋白。方法构建MCM7全长cDNA-pGBKT7质粒,与含肺癌组织cDNA文库-pACT2质粒转化酵母AH109,在涂有X-a-gal营养缺陷型培养基(SD/-Trp-Leu-His-Ade)上筛选生长,挑选蓝色克隆,提取此酵母克隆的质粒,转化大肠杆菌提取质粒DNA后测序,进行生物信息学分析。结果筛选出11种与MCM7蛋白特异性相互作用的蛋白,其中细胞骨架蛋白5种,各种酶、激酶以及相关受体6种。结论克隆到的基因对进一步研究MCM7蛋白的功能有一定提示作用。 Background and objective MCM7 is a subunit of the MCM complex that plays a key role in DNA replication initiation. But little is known about its interaction proteins. In this study yeast two hybrid screening was used to identify the MCM7 interacting proteins.
