摘要
目的 探讨中药复方益气解毒颗粒抑制鼻咽癌细胞裸鼠移植瘤生长的分子机制。 方法 将鼻咽癌细胞系HNE1接种至BABL/c裸小鼠 ,待移植瘤生长至一定大小 (约 1cm× 1cm× 1cm) ,灌喂益气解毒颗粒药液 (按人口服药量换算鼠的给药量 ) ,一个疗程 ( 3 0d)后处死裸鼠 ,解剖获取肿瘤组织 ,测量瘤体体积大小 ,称量瘤体重量。取 10 0mg瘤体组织 ,提取总RNA后逆转录标记成cDNA探针 ,与 5 88个基因的AtlasTMHumanCancercDNAArray杂交 ,灰度扫描杂交信号及统计分析差异。 结果 益气解毒颗粒药液能明显抑制鼻咽癌细胞裸鼠移植瘤的生长 ,灌喂中药组瘤体积和重量分别为 ( 0 .2 0 7± 0 .0 2 3 )cm3 和 ( 0 .13 2± 0 .0 2 1)g ,对照组为 ( 1.3 42± 0 .2 98)cm3 和 ( 1.0 17± 0 .0 15 )g(P <0 .0 5 ) ,抑制率为 84.5 8%。HumanCancercDNAArray分析发现 ,益气解毒颗粒组有STK1、MAPK1、CDK1等 19个与细胞增殖、细胞周期、核转录因子、DNA修复相关基因表达下调。有Caspase、TNFR -1、FADD等与细胞凋亡、角化蛋白死亡受体相关基因表达上调 ,部分基因表达与未用药组组织基因表达无明显差异。 结论 益气解毒颗粒抑制鼻咽癌裸鼠移植瘤的形成 ,可能是通过抑制细胞内重要的核转录因子、细胞分裂增殖、细胞周期、DNA修复和?
Objective To investigate the inhibitory mechanism of YIQIJIEDU granule on the growth of the implanted tumor of nasopharyngeal carcinoma cell line (HNE1) in nude mice. Methods 5×10 6 HNE1 cells were injected into BABL/c nude mice. Implanted-tumors grew for 20d,whose volume reached 1 cm×1 cm×1 cm. The nude mice were bred YIQIJIEDU granule for 30 d,whose growth was inhibited. The volume and weight of implanted-tumors was measured. The samples were obtained from implanted-tumors,which total RNA was extracted in for gene expression. Reserve transcription probes from the RNAs were respectively hybridized to the Atlas TM Human Cancer cDNA Array. Results The YIQIJIEDU granule could inhibit the growth of implanted-tumors. The inhibitory rate is 84.58%. In the YIQIJIEDU granule group,some genes in array were lowly expressed,such as STK 1,CDK,MAPK,et al. which associated with cell proliferation,cell cycle and nuclear factor,DNA repairing, Several genes in this group,such as caspase,TNFR-1,FADD and so on,which associated with apoptosis,keretin and death,were up-regulated. Conclusion YIQIJIEDU granule could inhibit the growth of nasopharyngeal carcinoma via blocking the signal pathway of cell proliferation and division,DNA repairing,cell cycle and inducing apoptosis and necrosis.
