摘要
目的从小鼠骨髓中体外诱导、扩增树突状细胞(DCs)。方法从小鼠骨髓中分离单个核细胞(MNCs),在rmGM-CSF、IL-4和TNF-α的诱导下培养扩增树突状细胞。光学显微镜观察其体外培养过程中的形态特征,流式细胞仪(FACS)分析其表型特征,混合淋巴细胞反应(MLR)检测其功能。结果MNCs经过rmGM-CSF、IL-4和TNF-α诱导培养12天后,具典型的DCs形态,细胞表型为MHCⅡ类分子(Ia)78.70±11.35%、CD11c81.60±10.02%、CD8065.30±5.73%、CD8679.65±8.66%、和CD1416.42±1.82%,具有极强的激发MLR的能力,显示成熟DCs的特征。结论本方法是一种可靠的诱导、扩增DCs的方法,为DCs的深入研究和临床应用奠定了基础。
Objective To induce and proliferate dendritic cells from murine bone marrow in vitro.Methods The mononuclear cells (MNCs) isolated from murine bone marrow were cultured in RPMI Medium 1640 containing rmGM-CSF,IL-4,and TNF-α to generate dendrite cells.The morphologic characteristics of these cells were observed under light microscopes and the phenotypic figures were analyzed with FACS.The function of these cells was reflected in mixed leukocyte reaction assay (MLR).Results The mononuclear cells (MNCs) were cultured in present of rmGM-CSF,IL-4,and TNF-α for 12 days,these cells bear typical morphologic and phenotypic characteristics of dendrite cells,expressed MHCⅡ(Ia) 78.70±11.35%,CD11c 81.60±10.02%,CD80 65.30±5.73%,CD86 79.65±8.66% and CD14 16.42±1.82%,gained the powerful capacity to stimulate allogeneic T cells.They showed the characteristics of mature DCs.Conclusions It is a reliable method to induce and proliferate DCs,which may facilitate further studies of DCs and its clinical application.
