摘要
目的:探讨牛初乳类胰岛素生长因子-I(IGF-I)对大鼠高血糖的治疗作用及其机理。方法:运用链脲佐菌素(STZ)建立糖尿病大鼠模型后,给予模型大鼠一定剂量的IGF-I,口服组连续30d,腹腔注射组连续15d;各组每隔5d测定血糖值,并于0d、15d、停药15d测定血清胰岛素、血清生长激素和血清游离IGF-I;实验结束后,取各组大鼠的胰腺分别进行病理切片观察以及免疫组织的化学分析。结果:实验结束时与糖尿病对照组比较,口服组血糖值和血清生长激素均显著下降(P<0.05,P<0.01),血清胰岛素水平无差异(P>0.05),血清游离IGF-I显著升高(P<0.05);腹腔注射组血糖值和血清生长激素均极显著降低(P<0.01),血清胰岛素和血清游离IGF-I均显著升高(P<0.05,P<0.01)。胰腺组织切片及免疫组织化学染色分析表明,口服组和腹腔注射组大鼠的胰腺形态以及β-细胞数量较糖尿病对照组均有显著的改善。结论:牛初乳IGF-I对大鼠高血糖有一定的治疗作用,其机理为保护胰腺β-细胞免受STZ损伤,并在一定程度上促进其增殖。
Objective:To study the effect and mechanism of bovine colostrums IGF-I on hyperglycemia in STZ induced diabetic rats.Methods:The STZ was injected intraperitoneally to the rats to establish diabetic animal model.Then 15 μgIGF-I/kg were administered by oral to diabetic rats for 30 d,10 μgIGF-I/kg by intraperitoneal injection for 15 d.Blood glucose were monitored at every 5 d and serum insulin,serum growth hormone and serum free IGF-I at every 15 d.The pathologic dyeing and insulin immunohistochemical slice of rats pancreas were observed and analyzed.Result:Compared with diabetic control rats,blood glucose and serum growth hormone level of oral group rats were decreased significantly(P<0.05,P<0.01),serum insulin level were no difference(P>0.05)and serum free IGF-I were increased significantly(P<0.05).While blood glucose and serum growth hormone level of intraperitoneal injection group rats were remarkably lower(P<0.01),serum insulin and serum free IGF-I level were remarkably higher(P<0.05,P<0.01)than that in diabetic control rats.The pathologic and insulin immunohistochemical slice showed that the pancreas conformation and the number of β-cell were all obviously improved than diabetic control rats.Conclusion:bovine colostrums IGF-I could improve hyperglycemia in STZ-induced diabetic rats and the mechanism were to protect the pancreas β-cell from STZ and promote the regeneration of pancreas β-cell to some extent.
