摘要 以刺五加的芽为外植体,研究不同消毒时间(3、5、7、10 min)、不同浓度的生长调节物质(6-BA、NAA、IBA)对芽诱导和增殖的影响。试验结果表明:刺五加外植体用75%乙醇消毒处理30 s,再用0.1%升汞消毒,其最佳时间为7min,外植体污染率低,存活率高。刺五加的最佳初分化培养以MS为培养基,最佳激素组合为6-BA 1.5 mg·L-1+NAA 0.5 mg·L-1,芽诱导率为85.56%;增殖培养以MS为培养基,最佳激素组合为6-BA1.0mg·L-1和IBA0.5 mg·L-1,芽增殖3.4倍,且生长健壮。 The effects of different disinfection time(3,5,7,10 min)and different concentrations of growth regulators(6-BA,NAA,IBA)on bud induction and proliferation of Acanthopanax senticosus were studied with the buds of Acanthopanax senticosus as explants.The results showed that the best disinfection method for explants was 75%ethanol disinfection for 30 s,and 0.1%mercuric chloride disinfection for 7 min,the contamination rate of explants was low and the survival rate was high.The optimal primary differentiation medium of Acanthopanax senticosus was MS medium,and the optimal hormone combination was 6-BA 1.5 mg·L-1+NAA 0.5 mg·L-1,and the bud induction rate was 85.56%;MS was used as the medium for proliferation culture,and the optimal hormone combination was 6-BA 1.0 mg·L-1+IBA 0.5 mg·L-1,and the bud proliferation multiple was 3.4 and the growth was robust.
