摘要
以中国石竹为试材,将pBI121-GUS分别转化在农杆菌GV3101和EHA105中,采用农杆菌侵染的方法侵染中国石竹离体花苞,研究了侵染后花苞绽放率和GUS染色程度,以期建立基因瞬时过表达体系,进而为将来快速筛选对花色表达有影响的基因提供参考依据。结果表明:农杆菌GV3101侵染后,花苞绽放率为56.67%,比EHA105的高15.84%;农杆菌侵染液OD_(600)值在1.4±0.2时,GUS染色效果最明显。因此,采用侵染液浓度为1.4±0.2的GV3101进行侵染时,可获得明显的中国石竹基因瞬时过表达结果。
D.chinensis was used as test materials.The vector pBI121-GUS was transformed into Agrobacterium tumefaciens GV3101 and EHA105,respectively.After the detached flower buds of D.chinensis infected,the flower bud blooming rate and GUS staining were studied,in order to provide reference for the rapid screening of genes aftecting the expression of flower color.The results showed that after infection with Agrobacterium GV3101,the flower bud blooming rate was 56.67%,which was 15.84%higher than that of EHA105.The color of GUS staining was obvious when the OD_(600)of the infiltration solution was 1.4±0.2.In conclusion,Agrobacterium GV3101 with its OD600of 1.4±0.2 should be better to be used in the transient overexpression system of D.chinensis.
