摘要
目的检测电针干预后神经病理性疼痛大鼠LC3-II蛋白表达水平的变化以探讨其对自噬的影响。方法60只SD大鼠随机分为5组(每组各12只):正常组、假手术组、膀胱经电针组、胆经电针组、模型组。除正常组和假手术组外,余3组建立坐骨神经结扎(CCI)模型。各组大鼠于术前(0天)及术后第3、14、21天均测定机械性刺激缩足阈值(PWMT)和热痛缩足反应潜伏期(PWTL);术后第14、21天采用Western Blot法测定坐骨神经及背根神经节内LC3-II蛋白表达水平。结果术前,各组大鼠PMWT和PWTL差异无统计学意义;术后第3天,模型组、胆经电针组、膀胱经电针组PMWT和PWTL均低于正常组和假手术组,说明造模成功;术后第14天,与模型组相比,电针组PMWT和PWTL均升高(P<0.05);术后第21天,电针组PMWT和PWTL持续升高,且胆经电针组增幅较膀胱经电针组大(P<0.05)。术后第14、21天,背根神经节内LC3-II的表达量各组间差异无统计学意义(P<0.05);术后第14天,相比模型组而言,胆经电针组大鼠坐骨神经内LC3-II的表达量降低(P<0.05),而膀胱经电针组与之无显著性差异(P<0.05);术后第21天,与模型组比较,膀胱经电针组大鼠坐骨神经内LC3-II表达降低不显著(P<0.05);与模型组、膀胱经电针组比较,胆经电针组坐骨神经内LC3-II的表达降低,差异有统计学意义(P<0.01)。结论电针足少阳胆经可上调CCI大鼠PWMT及PWTL,减少自噬关键因子LC3-II的表达量,提示电针治疗神经病理性疼痛可通过调节自噬而参与镇痛过程。
Objective To investigate the effect of electroacupuncture on autophagy by detecting the expression of LC3-II protein in neuropathic pain rats.Methods Sixty SD rats were randomly divided into 5 groups(12 in each group):normal group,sham operation group,bladder meridian electroacupuncture group,gallbladder meridian electroacupuncture group and model group.In addition to the normal group and the sham operation group,the remaining 3 groups established sciatic nerve ligation(CCI)models.Rats in each group were measured before operation(0 day)and 3,14,21 days after operation paw withdrawal mechanical threshold(PWMT)and paw withdraw thermal latency(PWTL).The expression of protein in sciatic nerve and spinal dorsal root(DRG)was determined by western blot method on day 14 and 21.Results Before operation,there was no significant difference in PMWT or PWTL rats in each group.On 3 rd day after operation,PMWT and PWTL in model group,gallbladder meridian electroacupuncture group and bladder meridian electroacupuncture group were lower than those in normal group and sham operation group,indicating that the model was successfully established.On the 14 th day after operation,PMWT and PWTL in electroacupuncture group were significantly higher than those in model group(P<0.05).On the 21 st day after operation,PMWT and PWTL in the electroacupuncture group increased continuously,and the increase in the gallbladder meridian electroacupuncture group was larger than that in the bladder meridian electroacupuncture group(P<0.05).There was no significant difference in the expression of LC3-II in the dorsal root ganglion of spinal cord on the 14 th and 21 st day after operation(P<0.05).On the 14 th day after operation,compared with that of the model group,the expression of LC3-II in the sciatic nerve of the rats in the gallbladder meridian electroacupuncture group was significantly decreased(P<0.05),but there was no significant difference in the bladder meridian electroacupuncture group.On the 21 st day after operation,compared with that of the model group,the expression of LC3-II in the sciatic nerve of rats in the bladder meridian electroacupuncture group was not significantly reduced(P<0.05).Compared with that of the model group and the bladder meridian electroacupuncture group,the expression of LC3-II in the sciatic nerve of the gallbladder meridian electroacupuncture group was significantly decreased,and the difference was statistically significant(P<0.01).Conclusion Electroacupuncture at gallbladder meridian can up-regulate PWMT and PWTL in CCI rats,reduce the expression of LC3-II which is a key autophagy factor,suggesting that electroacupuncture treatment of neuropathic pain may participate in the analgesic process by regulating autophagy.
