摘要
目的:考查离体大鼠肠道菌群对射干饮片中异黄酮苷代谢转化的影响。方法:将射干饮片与离体大鼠肠道菌群在厌氧条件下孵育24 h,孵育后饮片用水清洗干净并干燥至干,采用HPLC法对射干饮片中射干苷和野鸢尾苷及代谢产物鸢尾黄素和野鸢尾黄素进行定量分析。结果:大鼠肠道菌群对射干饮片中射干苷和野鸢尾苷有显著的代谢转化作用,孵育24 h时有79%射干苷和70%野鸢尾苷被转化,代谢产物分别为鸢尾黄素和野鸢尾黄素。结论:离体大鼠肠道菌群对射干饮片中的异黄酮苷类成分有代谢转化作用,而这种作用显著提高了射干饮片异黄酮苷元的含量。
Objective:To study the metabolic transformation of isoflavone glycoside in Rhizoma Belamcandae by rat intestinal flora in vitro.Method:Rhizoma Belamcandae was used for anaerobic incubation with rat intestinal flora in vitro for 24 h,then to be cleaned and dried.HPLC was used to detect the changes of isoflavone.Result:Tectoridin and Iridin could be easily metabolizedby rat intestinal flora in vitro.About 79%and 70%of tectoridin and Iridin were metabolized in 24 h,The major metabolite were tectorigenin and irigenin,respectively.Conclusion:The intestinal flora of isolated rats had a metabolic transformation effect on isoflavone glycosides in Rhizoma Belamcandae,the contents of isoflavones in Rhizoma Belamcandae were increased by metabolic.
