摘要
目的探讨环状RNA ASAP1(circ ASAP1)、微小RNA-338-3p(miR-338-3p)在结直肠癌(CRC)中的表达水平及其临床意义。方法选取2016年1月-2018年10月青岛市市立医院收治的78例CRC患者进行研究,收集所有CRC患者手术切除的CRC组织及对应的癌旁组织,测定CRC组织及对应的癌旁组织中circ ASAP1、miR-338-3p表达水平;分析CRC组织circ ASAP1、miR-338-3p表达水平与CRC患者临床病理特征、预后的关系;分析CRC组织中circ ASAP1表达水平与miR-338-3p的相关性。结果与癌旁组织比较,CRC组织circ ASAP1[(1.01±0.34)vs.(2.45±0.82)]表达水平较高,miR-338-3p表达水平[(1.04±0.35)vs.(0.49±0.17)]较低,差异有统计学意义(t=14.327、12.484,P均<0.05)。CRC组织circ ASAP1、miR-338-3p表达水平与肿瘤TNM分期、分化程度、淋巴结转移相关(P均<0.05)。CRC组织中circ ASAP1表达水平与miR-338-3p成负相关(r=-0.575,P<0.05)。circ ASAP1高表达组、miR-338-3p低表达组CRC患者累积生存率分别低于circ ASAP1低表达组、miR-338-3p高表达组(P均<0.05)。结论CRC组织中circ ASAP1表达水平较高,miR-338-3p表达水平较低,二者成负相关,且circ ASAP1、miR-338-3p有望成为临床诊治CRC、评估CRC预后的辅助指标。
Objective To investigate the expression levels and clinical significance of circular RNA ASAP1(circ ASAP1)and microRNA-338-3p(miR-338-3p)in colorectal cancer(CRC).Methods A total of 78 CRC patients accepted by our hospital from January 2016 to October 2018 were enrolled for the study.The surgically resected CRC tissues and corresponding paracancerous tissues of all CRC patients were collected.The expression levels of circ ASAP1 and miR-338-3p in CRC tissues and corresponding paracancerous tissues were determined;the relationship between the expression levels of circ ASAP1 and miR-338-3p in CRC tissues and the clinicopathological characteristics and prognosis of CRC patients was analyzed;the correlation between the expression levels of circ ASAP1 and miR-338-3p in CRC tissues was analyzed.Results Compared with adjacent tissues,the expression level of circ ASAP1(1.01±0.34)vs.(2.45±0.82)in CRC tissues was higher,and the expression level of miR-338-3p(1.04±0.35)vs.(0.49±0.17)was lower(t=14.327,12.484,all P<0.05);the expression levels of circ ASAP1 and miR-338-3p in CRC tissue were correlated with tumor TNM stage,degree of differentiation,and lymph node metastasis(P<0.05);the expression level of circ ASAP1 in CRC tissue was negatively correlated with miR-338-3p(r=-0.575,P<0.05);the cumulative survival rates of CRC patients in circ ASAP1 high expression group and miR-338-3p low expression group were lower than those in circ ASAP1 low expression group and miR-338-3p high expression group(all P<0.05).Conclusion The expression level of circ ASAP1 in CRC tissue was high,and the expression level of miR-338-3p was low,and the two were negatively correlated,and circ ASAP1 and miR-338-3p were expected to become auxiliary indicators for clinical diagnosis and treatment of CRC and evaluation of CRC prognosis.
