多重PCR检测技术在儿童急性呼吸道病毒感染中的应用 被引量:4

摘要 目的探讨多重PCR检测技术在儿童急性呼吸道感染中的临床应用价值。了解重庆市九龙坡区儿童急性呼吸道病毒感染病原情况,有助于更好地制定有效的预防策略。方法收集2021年3月-2022年2月重庆地区195例儿童急性呼吸道病毒感染病例咽拭子标本,应用18种呼吸道病原体核酸实时荧光PCR检测试剂盒进行检测。结果195份样本中有143份(73.33%)检测出病毒感染阳性,其中119份为单一病毒感染,24份为多重病毒感染。单一感染中最常见的是呼吸道合胞病毒25.64%(50/195),其次是人鼻病毒17.95%(35/195)、甲型流感病毒4.62%(9/195)。以0~3岁组阳性检出率最高为84.95%(79/93),>3~6岁组阳性检出率为60.00%(39/65),>6岁组阳性检出率为67.57%(25/37),不同年龄组病毒阳性检出率差异有统计学意义(χ^(2)=12.952,P=0.002)。冬春季节比夏秋季节更容易发生病毒感染(75.68%vs.41.67%)(χ^(2)=23.252,P<0.001)。结论多重PCR技术对于快速诊断呼吸道多重病原体感染有重要的临床意义,有助于全面了解儿童急性呼吸道病毒感染的病原和混合感染情况。 Objective To explore the clinical application value of multiplex PCR detection technology in the detection of acute respiratory tract infection in children.Understanding the pathogens of acute respiratory virus infection cases in children in Jiulongpo district of Chongqing will help to better formulate effective prevention strategies.Method Throat swab specimens from 195 children with acute respiratory virus infection in Chongqing from March 2021 to February 2022 were collected,and 18 respiratory pathogen nucleic acid real-time fluorescent PCR detection kits were used for detection.Results Of the 195 samples,143(73.33%)tested positive for virus infection,119 of them were single virus infection,and 24 were multiple virus infection.The most common single infection was respiratory syncytial virus 25.64%(50/195),followed by human rhinovirus 17.95%(35/195)and influenza A virus 4.62%(9/195).The highest positive detection rate was 84.95%(79/93)in the 0-3 years group,60.00%(39/65)in the>3-6 years group,and 67.57%(25/37)in the>6 years group.There were statistical differences in the positive detection rate of virus among different age groups,and virus infection was more likely to occur in winter and spring than in summer and autumn(75.68%vs.41.67%,χ^(2)=23.252,P<0.001).Conclusion Multiplex PCR technology had important clinical significance for the rapid diagnosis of multiple pathogen infections in the respiratory tract,and could be helpful for a comprehensive understanding of the pathogens and mixed infections of acute respiratory viral infections in children.
