基于“脾主肌肉”探讨葛根复方对实验性重症肌无力大鼠骨骼肌PGC-1α蛋白的影响 被引量:5

Effect of Pueraria Lobata Compound on PGC-1α Proteinin Skeletal Muscle for Rats with Experimental Myasthenia Gravis Based on the Theory of “Spleen Governs Muscle”

摘要 目的:探讨葛根复方对实验性重症肌无力大鼠(EAMG)骨骼肌PGC-1α蛋白的影响。方法:将80只SPF级雌性大鼠中随机选取10只留作空白组,其余的大鼠分别在后趾蹼、腹部、背部注射人工鼠源性AchR-α亚基97-116肽段乳化剂进行EAMG模型构建。模型构建成功后将剩余的70只大鼠随机分为模型组、葛芪组、葛根组、葛参组、葛芪参妙组、葛妙组、强的松组,共7组。每天给予葛芪组(3.65g/kg)、葛根组(2.08g/kg)、葛参组(3.65g/kg)、葛芪参妙组(11.46g/kg),葛妙组(8.33g/kg)、强的松组(5.4mg/kg)相应的药物灌胃,空白组和模型组给予生理盐水(3ml)灌胃,每日1次,连续28d,期间每两周测一次体重。治疗结束后,使用ELSIA法检测大鼠腹主动脉血清中AchR-ab含量;使用Western blot法检测各组大鼠骨骼肌组织中PGC-1α蛋白表达。结果:与模型组相比,葛根复方组和强的松组能够改善大鼠的体质量及降低AchR-ab的含量(P<0.05),提高PGC-1α蛋白的表达(P<0.05),其中葛芪参妙效果更明显(P<0.01)且优于强的松组(P<0.05)。结论:葛根复方可提高重症肌无力大鼠骨骼肌中PGC-1α蛋白的表达,从而对重症肌无力大鼠起到治疗效果。 Objective:To investigate the effect of Pueraria lobata compound on PGC-1α protein in skeletal muscle for(EAMG)rats with experimental myasthenia gravis.Methods:Ten of 80 SPF grade female rats were randomly selected as blank group.The other rats were injected with artificial AchR-α subunit 97-116 peptide emulsifier into the posterior toe web,abdomen and back to construct EAMG model.After successful replication of the model,the residual 70 rats were randomly fall into a model group,a Pueraria lobata group,a Puqi Shenmiao group and a prednisone group.The Pueraria lobata group(2.08 g/kg),the Puqi group(3.65 g/kg),the Pueraria group(3.65 g/kg),the Puxiao group(8.33 g/kg),the Puqi Shenmiao group(11.46 g/kg),the prednisone group(5.4 mg/kg)were given normal saline(3 ml)once a day for 28 days,and the body weight was measured every two weeks.At the end of treatment,the content of AchR-ab in the serum of abdominal aorta of rats was detected by ELSIA;the expression of PGC-1α protein in the skeletal muscle tissue of rats in each group was detected by Western blot.Results:Compared with the model group,the Pueraria lobata compound group and the prednisone group could improve the body mass,decrease the content of AchR-ab and increase the expression of PGC-1αprotein(P<0.05),and the effect of the Puqi Shenmiao group was more obvious than that of the prednisone group(P<0.05).Conclusion:Pueraria lobata compound can increase the expression of PGC-1α protein in skeletal muscle of myasthenia gravis rats,and thus has therapeutic effect on myasthenia gravis rats.
