摘要
目的:研究活血解毒方对放射性肺损伤小鼠模型肺组织的保护作用;方法:雄性C57BL/6小鼠108只,随机分为空白对照组、照射模型组、阳性药注射用甲泼尼龙琥珀酸钠腹腔注射组(0.05g·kg^(-1))、活血解毒方高、中、低剂量干预组(分别为13.9,6.96,3.48g·kg^(-1)),共6组,每组18只,各组小鼠均于造模前一周连续给药至造模后取材结束,其中空白对照组、照射模型组每日给予等量蒸馏水灌胃。采用6MV-X射线,经前胸野单次照射小鼠肺脏中平面18Gy,于照射结束后72h、第7天、第14天各组分别随机取小鼠断颈处死,取双肺标本,左肺用4%多聚甲醛固定2天后包埋,右肺分成3块后置-80℃保存。左肺组织常规苏木精-伊红(Hematoxylin and eosin,HE)染色,光镜下观察病理组织结构;酶联免疫法(Elisa)检测肺组织肿瘤坏死因子-α(Tumor Necrosis Factor alpha,TNF-α)、白介素-6(Interleukin-6,IL-6)的表达;通过实时荧光定量PCR技术对肺组织转化生长因子-β1信使RNA(Transforming growth factor-β1messenger RNA,TGF-β1mRNA)进行定量分析。结果:(1)活血解毒方中剂量可降低照射诱导肺损伤小鼠造模后72h、第7天肺损伤病理评分;活血解毒方低及高剂量组在造模后第7天肺损伤病理评分显著降低。(2)活血解毒方中剂量可显著降低造模后第7天TNF-α表达;活血解毒方高剂量组可明显下调第14天TNF-α表达。(3)活血解毒方高剂量组可显著降低造模后第7天TGF-β1mRNA表达;活血解毒方高及中剂量均可明显下调第14天TGF-β1mRNA表达。结论:活血解毒方对放射性肺损伤小鼠模型肺组织具有一定的保护作用。
Objective:To study the protective effect of Huoxue Jiedu Recipe on lung tissue for mice with radiation lung injury.Methods:108 male C57 BL/6 mice were randomly divided into a blank controlled group,an irradiation model group,an intraperitoneal injection group of methylprednisolone sodium succinate for positive drug injection(0.05 g·kg^(-1)),high blood,medium and low blood circulation and detoxification.The dose intervention group(13.9,6.96,3.48·kg^(-1),respectively),a total of 6 groups,18 in each group.All groups of mice were administered continuously from the week before the moulding to the end of the moulding,in which the blank control group and the irradiation model group were given the same amount of distilled water daily.The 6 MV-X-rays were used to irradiate the lungs in the lungs with a single plane of 18 Gy.The rats were randomly sacrificed at 72 h,7thand 14thafter the end of the irradiation,and the lung specimens were taken.The left lung was fixed with 4%paraformaldehyde for 2 days,and the right lung was divided into 3 pieces and stored at-80°C.The left lung tissue was stained with Hematoxylin and eosin(HE).The pathological structure was observed under light microscope.The expression of TNF-αand IL-6 in lung tissue was detected by enzyme-linked immunosorbent assay(ELISA).Quantitative analysis of lung tissue TGF-β1 mRNA by quantitative PCR.Results:(1)The medium dose of Huoxue Jiedu Decoction could reduce the pathological score of lung injury at 72 h and 7 days after modeling in mice with radiati-oninduced lung injury.The pathology of lung injury on the 7thday after model establishment in Huoxue Jiedu Recipe and high dose group.The score is significantly lower.(2)The medium dose of Huoxue Jiedu Recipe could significantly reduce the expression of TNF-αon the 7thday after model establishment,the high dose group of Huoxue Jiedu Recipe could significantly down-regulate the expression of TNF-αon the 14thday.(3)The high-dose group of Huoxue Jiedu Recipe could significantly reduce the expression of TGF-β1 mRNA on the 7thday after model establishment,the high and medium doses of Huoxue Jiedu Recipe could significantly down-regulate the expression of TGF-β1 mRNA on the 14thday.Conclusion:Huoxue Jiedu Decoction has a certain protective effect on lung tissue of mice with radiation-induced lung injury.
