铜-还原碘量法测定薏苡仁可溶性糖含量 被引量:9

Determination the Content of Soluble Sugar in Coix Seed by Shaffer-Somogyi

摘要 建立薏苡仁可溶性糖含量的测定方法,单因素试验优化可溶性糖最佳提取条件。以葡萄糖为标准品,采用铜还原碘量法测定薏苡仁可溶性糖的含量。结果表明,最佳提取条件为:5 g薏苡仁细粉,置250 mL锥形瓶中,加提取溶剂水100 mL,置于45℃水浴中振荡提取60 min。该方法的线性范围为0~2.5 mg,回归方程为y=4.902 9x+0.238 1,R^2=0.998 8,平均回收率为91.6%(RSD=1.7%, n=9),精密度RSD值为2.5%,稳定性RSD=1.6%,检出限0.2 mg/g。该方法灵敏、稳定、重现性好,可用于薏苡仁可溶性糖含量的测定。 To establish a determination method of soluble sugar in Coix seed, the optimum extract determination conditions was investigated by single factor experiments. The contents of soluble sugar was measured by Shaffer-Somogyi method. And these test used glucose as the standard substance. The optimum determination conditions were as follows: 5 g Coix seed powder, 250 mL container, extract solution 100 mL, and 45 ℃ heated water for 60 min. The linear range of this method was 0-2.5 mg, and the regression equation was y=4.902 9 x+0.238 1, R^2=0.998 8, and the average recovery rate was 91.6%(RSD=1.7%, n=9), accuracy RSD under the value 2.5%, stable RSD=1.6%, and detection limit 0.2 mg/g. This method was sensitive and stable, which could be used to determine the contents of soluble sugars in Coix seed with good reproducibility.
