摘要
目的综述甾体化合物羟基化修饰的生物合成研究进展,为解决甾体衍生物溶解度低和合成困难等问题,提供新思路。方法总结近几年国内外发表的29篇论文,将甾体化合物采用生物转化法进行羟基化修饰过程研究进展进行分类、归纳并总结。结果细菌和丝状真菌都可以对不同的甾体化合物进行不同位点的羟基化,其中以丝状真菌为主。通过不同的微生物对甾体进行羟基化修饰可以增大甾体化合物的溶解度,增加药效或降低毒性以及提高生物利用度。结论生物转化法利用微生物体内的生物酶可以在一步之内得到不同的甾体衍生物,为绿色快捷地合成种类丰富的甾体类化合物提供了参考,为最终实现临床应用奠定了基础。
Objective To review the progress in the biosynthesis of steroid derivatives modified by hydroxylation,and to provide a new way to solve the problems of low solubility and difficult synthesis of steroid derivatives.Methods Twenty-nine papers published in recent years were summarized,and the research progress of steroid hydroxylation by biotransformation was classified,deduced and summarized.Results Bacteria and filamentous fungi is capable of hydroxylating different steroids at different sites,among which filamentous fungi is the main one.Hydroxylation of steroids by different microorganisms can increasing the solubility of steroids,increasing the efficacy or reduce the toxicity,and improving the bioavailability.Conclusion Different steroidal derivatives can be obtained in one step by biotransformation,which provides a reference for the green and rapid synthesis of various steroids and lays a foundation for the final clinical application.
