摘要
目的:研究8种壮瑶药提取物含药血清对肝星状细胞增殖的影响。方法:健康家兔18只,随机分为9组,对照组,8个壮瑶药提取物组(火炭母提取物、犁头草提取物、青钱柳提取物、拳卷地钱提取物,美丽胡枝子提取物、蝙蝠草提取物、锡叶藤提取物、白背三七多糖提取物,采用同一剂量40 g/kg)。对照组家兔按10 mL/kg灌胃给予纯净水,壮瑶药提物组灌胃相同浓度的壮瑶药提取物(0.4 g/mL,10 mL/kg),每天1次,连续给药6 d。采血并分离获得8种壮瑶药提取物的含药血清。采用结晶紫染色法测定8种壮瑶药提取物20%含药血清的细胞毒性,经共培养48 h后,用肉眼观察和拍照记录,测定肝星状细胞T6的吸光度值(A值),并计算细胞生长抑制率。结果:与对照组相比,8种壮瑶药提取物20%含药血清共培养48 h后,肝星状细胞T6数目明显减少,A值显著降低(P<0.01),细胞生长抑制率显著提高(P<0.01),8种壮瑶药提取物按对肝星状细胞T6生长抑制率由高到低排列依次为白背三七多糖(65.04%)、美丽胡枝子提取物(56.63%),锡叶藤提取物(43.03%)、犁头草提取物(41.67%)、蝙蝠草提取物(35.21%),拳卷地钱提取物(34.37%),青钱柳提取物(22.89%)和火炭母提取物(15.81%)。结论:8种壮瑶药提取物含药血清对肝星状细胞T6均有较好的生长抑制作用,其中白背三七多糖和美丽胡枝子提取物被认为作用效果最佳。
Objective:To study the effect of drugcontaining serum of eight extracts from zhuang and yao medicine on proliferation of hepatic stellate cells.Methods:Eighteen healthy rabbits were randomly divided into nine groups,control group,eight extracts from zhuang and yao medicine groups(Polygonum chinense L.extract,Viola inconspicua Bl.extract,Cyclocarya paliurus(Batal.)Iljinskaja extract,Marchantia convolute extract,Lespedeza formosa(Vog.)Koehne.Extract,Christia vespertilionis(L.f.)Bahn.f.extract,Tetracera asiatica(Lour.)Hoogland extract,Gynura divaricate polysaccharide,with the same dose 40 g/kg).Rabbits in control group were intragastrically administered with pure water(10 mL/kg),rabbits in extracts from zhuang and yao medicine groups were intragastrically administered with the same concentrations(0.4 g/mL,10 mL/kg)of extracts from zhuang and yao medicine,once a day for six consecutive days.Subsequently,their arterial blood collected and the drug-containing serum of eight extracts from zhuang and yao medicine separated.Crystal Voilet Staining was used to measure the cytotoxicity of 20%drug-containing serum of eight extracts from zhuang and yao medicine,after incubated for 48 h,visual examination and photo taking used for examining and recording,the A value of hepatic stellate cells T6 determined,and growth inhibition ratio calculated.Results:Compared with control group,after treated with 20%drug-containing serum of eight extracts from zhuang and yao medicine for 48 h,the numbers of hepatic stellate cells T6 was obviously decreased,the A value of hepatic stellate cells T6 was significantly decreased(P<0.01),and the growth inhibition rates was significantly increased(P<0.01),the growth inhibition rates of hepatic stellate cells T6 in eight different extracts from zhuang and yao medicine from high to low were Gynura divaricate polysaccharide(65.04%),Lespedeza formosa(Vog.)Koehne.Extract(56.63%),Tetracera asiatica(Lour.)Hoogland extract(43.03%),Viola inconspicua Bl.extract(41.67%),Christia vespertilionis(L.f.)Bahn.f.extract(35.21%),Marchantia convolute extract(34.37%),Cyclocarya paliurus(Batal.)Iljinskaja extract(22.89%)and Polygonum chinense L.extract(15.81%).Conclusion:Drug-containing serum of eight extracts from zhuang and yao medicine has good growth inhibition effect on hepatic stellate cells T6,and Gynura divaricate polysaccharide,Lespedeza formosa(Vog.)Koehne.Extract were considered to be the best.
