衰老影响小鼠脊髓组织锌转运体表达及细胞凋亡的实验研究 被引量:3

Effects of aging on the expression of zinc transporter and apoptosis in mouse spinal cord tissues

摘要 目的探讨衰老对小鼠脊髓组织中锌转运体表达及细胞凋亡的影响。方法取材青年小鼠和老年小鼠脊髓组织,应用HE染色观察灰质神经元形态;通过免疫组织化学染色和体视学技术检测锌转运体家族成员ZnT1、Zn T3、ZnT6和ZnT10的表达变化;应用免疫印迹技术检测凋亡相关蛋白Bax和Bcl-2含量变化。结果HE染色结果显示,老年小鼠脊髓灰质中神经元数量明显少于青年小鼠。免疫组织化学染色结果显示,Zn T1、Zn T3、ZnT6和ZnT10均表达于脊髓神经元;体视学测量结果显示,与青年小鼠相比,老年小鼠脊髓组织中ZnT1、ZnT3和ZnT6的表达呈不同程度的减弱,Zn T10在老年小鼠脊髓内的表达高于青年小鼠。免疫印迹结果表明,与青年小鼠比较,老年小鼠脊髓组织中抗凋亡蛋白Bcl-2的表达显著降低,促凋亡蛋白Bax的表达明显升高。结论衰老影响脊髓组织中锌转运体的表达,可能是老年小鼠脊髓组织神经元凋亡增加的原因之一。 Objective To explore the effect of aging on the expression of zinc transporter and cell apoptosis in mouse spinal cord tissue.Methods The spinal cord tissue of young and old mice were taken,HE staining was used to observe the morphology of neurons in spinal cord gray matter.The expressions of ZnT1,ZnT3,ZnT6 and Zn T10 were detected byimmunohistochemistryand stereological analysis.Western blot was used to detect the contents of apoptosis-related proteins Bax and Bcl-2 in the spinal cord of young and old mice.Results HE staining result showed thatthe number of neurons in the spinal cord gray matter of old mice was significantly less than that in young mice.The results of immunohistochemical staining and stereological analysis showed that Zn T1,Zn T3,Zn T6 and ZnT10 were all expressed in spinal cord neurons.Compared with young mice,the expression of ZnT1,Zn T3 and ZnT6 in spinal cord tissue of old mice was weakened to various degrees.However,the expression of Zn T10 in the spinal cord of old mice was higher than that of young mice.The data of western blot indicated that compared with young mice,the expression of anti-apoptotic protein Bcl-2 in the spinal cord tissue of old mice was significantly reduced,but the level of pro-apoptotic protein Bax was obvious increased.Conclusions Aging affects the expression of zinc transporters in spinal cord,which may be one of the reasons for the increased apoptosis of neurons in old mice.
