摘要
目的探讨孤独症谱系障碍儿童血清miR-106b表达及其与炎性因子的相关性。方法选取本院2018年8月-2019年7月收治的105例无合并症的孤独症谱系障碍儿童为病例组,73例健康儿童为对照组。采用RT-PCR法及ELISA法检测血清miR-106b及炎性因子IL-2、IL-6、TNF-α水平。探讨Pearson相关分析探讨血清miR-106b水平与炎性因子的相关性,ROC曲线比较miR-106b、IL-2、IL-6、TNF-α对孤独症谱系障碍的诊断价值。结果病例组miR-106b、IL-2、IL-6、TNF-α水平高于对照组,差异均有统计学意义(P<0.05)。病例组中度组、重度组miR-106b、IL-2、IL-6、TNF-α水平高于轻度组(P<0.05),中度组miR-106b、IL-2、IL-6、TNF-α水平高于轻度组,随着病例组严重程度增加,miR-106b、IL-2、IL-6、TNF-α水平逐渐升高(P<0.05)。miR-106b与IL-2、IL-6、TNF-α正相关(P<0.05)。IL-2、IL-6、TNF-α三者的AUC相近,皆小于miR-106b;IL-2、IL-6、TNF-α诊断孤独症谱系障碍的灵敏度、特异度、阳性预测值、阴性预测值相近(χ2=1.098、1.148、0.987、1.685、1.548、2.012、1.658、1.549、1.874、1.598、1.489、1.698,P>0.05),皆小于miR-106b(χ2=15.698、13.547、10.547、12.655、8.698、12.589、9.969、10.548、8.699、9.998、12.587、13.587,P<0.05)。结论孤独症谱系障碍儿童血清miR-106b的高表达与患儿神经系统炎症反应密切相关;miR-106b诊断孤独症谱系障碍的灵敏度、特异度、阳性预测值、阴性预测值较高,可在临床上推广应用。
Objective:To investigate the effect of between psychological status and coping style,self-efficacy and quality of life in cancer patients.Methods:From August 2018 to July 2019,105 children with autism spectrum disorder without comorbidity were selected as the case group and 73 healthy children as the control group.Serum levels of miR-106b,inflammatory factors including IL-2,IL-6 and TNF-a were detected by RT-PCR and ELISA.Pearson correlation analysis was used to explore the correlation between serum levels of miR-106b and inflammatory factors,and ROC curve was used to compare the diagnostic value of miR-106b,IL-2,IL-6 and TNF-αfor autism spectrum disorder.Results:The levels of miR-106b,IL-2,IL-6 and TNF-αin the case group were higher than those in the control group,and the differences were statistically significant(P<0.05).The levels of miR-106b,IL-2,IL-6 and TNF-αin the moderate group and severe group were higher than those in the mild group(P<0.05),while the levels of miR-106b,IL-2,IL-6 and TNF-αin the moderate group were higher than those in the mild group.With the increase of the severity of the case group,the levels of miR-106b,IL-2,IL-6 and TNF-αgradually increased(P<0.05).miR-106b was positively correlated with IL-2,IL-6 and TNF-α(P<0.05).The AUC of IL-2,IL-6,IL-6,TNF-αwas similar,which all were smaller than that of miR-106b.The sensitivity,specificity,positive predictive value,positive predictive value,negative predictive value of IL-2,IL-6,TNF-αfor the diagnosis of autism spectrum disorder were similar(χ~2=1.098,1.148,1.148,0.987,1.987,1.685,1.685,1.548,1.548,2.012,1.658,1.549,1.549,1.874,1.598,1.489,1.698,P>0.05),which all were smaller than that of miR-106b(b(χ~2=15.698,13.547,10.547,12.655,8.698,12.589,9.969,10.548,8.699,9.998,12.587,13.587,P<0.05).Conclusion:The high expression of serum miR-106b in children with autism spectrum disorder is closely related to the inflammatory response of the nervous system of children.The sensitivity,specificity,positive predictive value and negative predictive value of miR-106b in the diagnosis of autism spectrum disorder are high,which can be popularized and applied in clinic.
