摘要
目的:探讨葛根芩连汤含药血清对胰岛素抵抗(IR)的脂肪细胞糖代谢调控机制。方法:将正常脂肪细胞作为空白对照组、IR-脂肪细胞分为模型对照组、罗格列酮10 mg/L组、25 g/kg葛根芩连汤含药血清5%、10%、15%组,干预IR细胞6 h,12 h,18 h,24 h,以葡萄糖消耗量为药效指标,采用高分辨液相色谱串联四极杆飞行时间质谱(UPLC-Q-TOF/MS)分析技术检测各组细胞内的代谢产物信号,通过Mass Profiler Professional(MPP)软件分析数据,找出生物标记物,进一步通过二级质谱结构鉴定潜在的生物标记物,找出葛根芩连汤含药血清对胰岛素抵抗脂肪细胞糖代谢调控的可能作用机制。结果:通过25 g/kg葛根芩连汤5%、15%、15%含药血清作用于IR细胞模型,葛根芩连汤5%含药血清在18 h时能显著提高IR-脂肪细胞葡萄糖消耗量,改善脂肪细胞胰岛素抵抗。发现6个与糖代谢相关的生物标记物,其中鉴定出的物质有2个(腺嘌呤和5′-甲硫腺苷)。结论:根据通路分析,推测葛根芩连汤含药血清可能会上调细胞中腺嘌呤、5′-甲硫腺苷的含量,从而调节细胞内的嘌呤代谢途径、半胱氨酸和蛋氨酸代谢途径,调节细胞中的糖代谢。
Objective:To investigate the regulatory mechanism of Gegen Qinlian Decoction-containing serum on glucose metabolism in insulin-resistant(IR)adipocytes.Methods:With normal adipocytes as the blank control,the IR adipocytes were divided into the model group,10 mg/L rosiglitazone group,and low-,medium-,and high-dose(5%,10%,and 15%)Gegen Qinlian Decoction-containing serum(25 g/kg)groups,and intervened for 6 h,12 h,18 h and 24 h respectively with glucose consumption as the pharmacodynamic index.The intracellular metabolite signals in each group were detected by ultra performance liquid chromatography/quadrupole time of flight tandem mass spectrometry(UPLC-Q-TOF/MS).The collected data were analyzed by Mass Profiler Professional(MPP)to find out biomarkers,followed by potential biomarker identification by mass spectrometry/mass spectrometry(MS/MS).Thus,the possible regulatory mechanism of Gegen Qinlian Decoction-containing serum on glucose metabolism in IR adipocytes was figured out.Results:After treatment of IR adipocytes with 5%,10%,and 15%Gegen Qinlian Decoction-containing serum(25 g/kg),it was found that 5%Gegen Qinlian Decoction-containing serum significantly enhanced the glucose consumption in IR adipocytes at 18 h and improved IR of adipocytes.Six potential biomarkers related to glucose metabolism were discovered,of which two were identified to be adenine and 5′-methylthioadenosine.Conclusion:According to pathway analysis,Gegen Qinlian Decoction-containing serum may elevate the contents of adenine and 5’-methylthioadenosine in cells to regulate the metabolic pathways of purine,cysteine and methionine in cells and glucose metabolism.
