The effect of pH values on the extracellular poly-saccharide secreted by Acidithiobacillus ferrooxidans was investigated in different phases of bacterial growth during chalcopyrite bioleach-ing. It is found that extracellular polysaccharide secretion from the cells attached to chalcopyrite is more efficiently than that of the free cells in the bioleaching solution. Three factors, pH values, the concentration of soluble metal ions, and the bacterial growth and metabolism, affect extracellular polysaccharide secretion in the free cells, and are related to the bacterial growth phase. Extracellular polysaccharide secretion from the attached cells is mainly dependent on the pH value of the bacterial culture.展开更多

Gas chromatography-mass spectrometry (GC-MS) w...This study is intended to explore the chemical differences of Acori Tatarinowii Rhizoma (ATR) samples collected from two habitats, Sichuan and Anhui provinces, China. Gas chromatography-mass spectrometry (GC-MS) was applied to establishing the quantitative chemical fingerprints of ATRs. A total of 104 volatile compounds were identified and quantified with the information of mass spectra and retention index (RI). Furthermore, least absolute shrinkage and selection operator (LASSO), a sparse regularization method, combined with subsampling was employed to improve the classification ability of partial least squares-discriminant analysis (PLS-DA). After variable selection by LASSO, three chemical markers,β-elemene, α-selinene and α-asarone, were identified for the discrimination of ATRs from two habitats, and the total classification correct rate was increased from 82.76% to 96.55%. The proposed LASSO-PLS-DA method can serve as an efficient strategy for screening marked chemical components and geo-herbalism research of traditional Chinese medicines.展开更多

The gold nanoparticles with an average diameter of 5...A fast and facile method of fabricating fiber-optic localized surface plasmon resonance sensors baseff on spherical gold nanoparticles was introduced in this study. The gold nanoparticles with an average diameter of 55 nm were synthesized via the Turkevich method and were then immobilized onto the surface of an uncladded sensor probe using a polydopamine layer. To obtain a sensor probe with high sensitivity to changes in the refractive index, a set of key optimization parameters, including the sensing length, coating time of the potydopamine layer, and coating time of the gold nanoparticles, were investigated. The sensitivity of the optimized sensor probe was 522.80 nm per refractive index unit, and the probe showed distinctive wavelength shifts when the refractive index was changed from 1.328 6 to 1.398 7. When stored in deionized water at 4 ℃, the sensor probe proved to be stable over a period of two weeks. The sensor also exhibited advantages, such as low cost, fast fabrication, and simple optical setup, which indicated its potential application in remote sensing and real-time detection.展开更多

Among these technical applications, nano-composite food packaging, which combines the NMs and conventional packaging, is at the forefront of applications in nanotechnologies, leading the whole industrial chain based on nanotechnologies with high speed development. Taking into account the status of the nano-composites applied in food packaging and present development trends, this review is focused on summarizing the basic applications of the nano-composites in food packaging as well as its risk assessment. The status summaries and the conclusions derived from this review are of benefit to manufacturers which produce the nano-composites used in food packaging, to general consumers and to governmental administration entities.展开更多
In an attempt to isolate and characterize peptides mimicking epitopes of metalloprotease and explore their immunological protection against Schistosoma japonicum (S. japonicum), polyclonal anti-metalloprotease sera ...In an attempt to isolate and characterize peptides mimicking epitopes of metalloprotease and explore their immunological protection against Schistosoma japonicum (S. japonicum), polyclonal anti-metalloprotease sera was prepared to screen a 12-mer random peptide library to isolate phages binding specially to antisera IgG. Then, phage ELISA, animal immunization, DNA sequencing, Western blotting and enzymatic activity neutralizing analysis were used to characterize the selected phage clones. All of ten randomly picked clones were shown to be positive. Five peptides of different amino acid sequences deduced from DNA sequences were obtained and two of them (peptides 2 and 3) could induce significant reduction (31.0% and 31.8%, respectively) in worm burden and high reduction (52.6% and 54.9%, respectively) in liver eggs per gram (LEPG), while, unexpectedly, others (peptides 1, 4 and 5) could not elicit enough protection against infection of S. japonicum. Peptides 2 and 3 could be recognized by S. japonicum infected mouse sera (IMS) and could elicit neutralizing Abs. The results show that peptides 2 and 3 are antigenic and immunogenic. A novel methodology that combines high performance liquid chromatography with photodiode-array detector (HPLC-DAD) coupled with second-order calibration method based on alternating trilinear decomposition (ATLD) a...A novel methodology that combines high performance liquid chromatography with photodiode-array detector (HPLC-DAD) coupled with second-order calibration method based on alternating trilinear decomposition (ATLD) algorithm was used in determination of the effective constituents such as costunolide and dehydrocostuslactone, in plasma sample and Chinese patent medicine Xiang Sha Yang Wei (XSYW) capsule. Complicated systems such as plasma and Chinese patent medicine which have intricate components are tedious to isolate and purify. The problem that chromatographic peaks are heavily overlapped among the analytes and interferents from the background matrices can be resolved, and the satisfactory quantification results have been gained with the help of the ATLD algorithm which utilized "mathematical separation" instead of partial "physical or chemical separation". Meanwhile, HPLC-MS/MS method was used to validate the accuracy of the proposed determination method.展开更多
Two Schistosomajaponicum vaccine candidate antigens Sj 31 and Sj 32, which have shown particular promise to induce protective immunity in mice, were used to immunize goats by using a DNA priming-protein boosting strat...Two Schistosomajaponicum vaccine candidate antigens Sj 31 and Sj 32, which have shown particular promise to induce protective immunity in mice, were used to immunize goats by using a DNA priming-protein boosting strategy in present work. DNA vaccine formulations of the two antigens (VRSj31 and VRSj32) were produced and injected intramuscularly twice at a 2-week interval and then recombinant proteins (rSj31 and rSj32) together with Freund Complete Adjuvant (FCA) were used to boost the goats. The experiment was repeated in different batche cercariae. A strong anamnestic antibody response was induced after boost. A significant reduction of liver egg counts and miracidial hatching was showed in both experiments. Significant protections against challenge infection were elicited with 31.6% of percentage reduction for worm recovery in the second experiment and 20.9% in the first experiment, respectively.展开更多
