Methods: Mature (4 months old) Wistar rats were intraperitoneally administered sodium arsenite at doses of 4, 5 or 6 mg-kg^-day1 for 26 days. Different varieties of germ cells at stage VII seminiferous epithelium cycle, namely, type A spermatogonia (ASg), preleptotene spermatocytes (pLSc), midpachytene spermatocytes (mPSc) and step 7 spermatids (7Sd) were quantitatively evaluated, along with radioimmunoassay of plasma follicle-stimulating hormone (FSH), lutuneizing hormone (LH), testosterone and assessment of the epididymal sperm count. Results: In the 5 and 6 mg/kg groups, there were significant dose-dependent decreases in the accessory sex organ weights, epididymal sperm count and plasma concentrations of LH, FSH and testosterone with massive degeneration of all the germ cells at stage VII. The changes were insignificant in the 4 mg/kg group. Conclusion: Arsenite has a suppressive influence on spermatogenesis and gonadotrophin and testosterone release in rats.展开更多

According to day 0.5, 1, 3, 6, 10, 25, 35 and 50 after local testicular heating, every group was divided into 8 subgroups: experimental subgroups (n=6) and control subgroups (n=4). DNA contents of various types of germ cells were observed with flow cytometry (FCM) in all groups. Results Compared with control groups, percentages of 4C cell (primary spermatocyte) in 0.5-35 d groups and percentages of 1C cell (spermatid and sperm) in 6-50 d groups significantly decreased in experimental groups (P〈0.05), and percentages of 2C cell (spermatogonium and second spermatocyte) in 3 -35 d experimental groups increased significantly after heating (P〈0.05). 4C:2C in all of 8 experimental groups and 1C:2C in 3-35 d experimental groups were down (P〈0. 05), and in 1 d experimental group 1C:4C was up after heating (P〈0.05). The content of Bax positive cells increased in all experimental subgroups (P〈0.01), the ratio of positive cells which had no significant difference (P〉0.05) except 6 d group decreased (P〈0.01 ). Redistribution of Bax from a cytoplasmic to perinuclear or nuclear localization could be observed after heating. Conclusions Expression of Bcl-2 would decrease and Bax would increase with redistribution in spermatogenic cells in rats after heating. Anxiety is an unpleasant state,which can critically decrease the quality of life is often accompanied by nervous behaviour and rumination.Niranthin is a lignan isolated from various Phyllanthus sources.The literature ...Anxiety is an unpleasant state,which can critically decrease the quality of life is often accompanied by nervous behaviour and rumination.Niranthin is a lignan isolated from various Phyllanthus sources.The literature survey on niranthin highlights wide ranges of the therapeutic potentials.In a present study,based on our previous investigations,we evaluated pure,isolated and characterized niranthin as an anxiolytic agent.The niranthin[6-[(2R,3R)-3-[(3,4-dimethoxyphenyl)methyl]-4-methoxy-2-(methoxymethyl)butyl]-4-methoxy-1,3-benzodioxole]was purchased from commercial source and further subjected for assessment of its anxiolytic potentials using popular animal models including Elevated plus-maze model/test(EPM)and Light&Dark Exploration test(L&D).GABA-A receptor mediation was evaluated by pretreating the mice with the GABA-A receptor antagonist Flumazenil before the EPM task.Molecular docking simulation studies(pdb id:4COF)carried out by Vlife QSAR software showed that niranthin(docking score:−62.1714 kcal/mol)have shown comparatively best docking score compared to the standard drug Diazepam(docking score:−63.1568 kcal/mol).To conclude,Niranthin has probable potential in the management of anxiety disorder.Our in-silico and in-vivo analysis(indirectly)indicated the plausible role of GABA mediation for anxiolytic activity.Although,these studies are preliminary,future in depth experimental explorations will be required to use Niranthin as anti-anxiety drug in near future.展开更多

Odorants generate different changes in the concentrations of oxy- hemoglobin. Our results suggest that NIRS technology might be useful in discriminating various odorants in a non-invasive manner using animals with a superb olfactory system.展开更多
Objective:To develop new biomarkers for radio-resistance in colorectal cancer(CRC)to improve radiotherapy efficacy at lower position and late phase of CRC patients.Methods:The human CRC cell lines were irradiated by a...Objective:To develop new biomarkers for radio-resistance in colorectal cancer(CRC)to improve radiotherapy efficacy at lower position and late phase of CRC patients.Methods:The human CRC cell lines were irradiated by a total dose of 40 Gy.RAN-seq screened differentially expressed genes(DEGs)related to radio-resistance.We orientated the genes which play a vital role in radioresistance by bio-informatics analysis.Ultimately,the genetic expressions were verified by qPCR.Results:3,823radio-resistance genes were found in RKO cells and 4,057 DEGs were in HCT116 cells,compared with the same type cells without radiation treatment.Data of enrichment analysis showed that DEGs obtained from two cell lines were mainly involved in cell cycle,cell division,cell skeleton,cell adhesion,DNA replication,DNA damage and DNA repair,and enrichment of glycometabolism,p53 and HIF-1 signaling pathways.The 10 candidate biomarkers were predicted according to the interaction with each other,and 5 genes(CD44,CXCL-8,ITGA5,ITGB8 and STAT1)were confirmed markedly up-regulated in two cell lines.Conclusion:Radio-resistance of CRC cells induced by X-irradiation changed the variety of genes expression,and those nodes among these genes network interaction may play core regulatory roles,which will provide new therapeutic strategies for clinical radiotherapy。展开更多
